摘    要:目的:建立以反相高效液相色谱法测定人血浆中烟酸浓度的方法。方法:以咖啡因为内标,血浆样品用10%硫酸锌沉淀后进行分析,色谱柱为DiamonsilTM C18,流动相为乙腈-0.01mol·L-1磷酸二氢钾(内含0.01mol·L-1四丁基溴化铵,pH6.5)=15:85,流速为1.0mL·min-1,检测波长为262nm,柱温为40℃。结果:烟酸血药浓度在0.2~20.0μg·mL-1范围内线性关系良好(r=0.9999),最低检测浓度为0.1μg·mL-1;低、中、高浓度的回收率在93.27%~102.65%之间,日内、日间RSD均<8%。结论:本方法准确、灵敏、重复性好,且不受他汀类药物的干扰,能为烟酸,尤其临床上与他汀类药物联合应用时的血药浓度监测及人体药动学研究提供检测手段。

Abstract:OBJECTIVE:To establish an RP-HPLC method for the concentration determination of niacin in human plasma.METHODS:Using caffeine as internal standard,the plasma sample was precipitated with 10%ZnSO 4.The separation was carried out on Diamonsil TM C 18 (250 mm×4.6 mm,5 μm)column with mobile phase consisted of acetonitrile-0.01 mol · L-1 KH 2 PO 4 buffer solution(pH=6.5,containing 0.01 mol · L-1 tetrabutyl ammonium bromide)(15:85).The flow rate was 1.0 mL · min-1.The detection wavelength was 262 nm.The column temperature was 40 ℃.RESULTS:The linear range was 0.2~20.0 μg · mL-1 (r=0.999 9).The lowest detection concentration was 0.1 μg · mL-1.The intra-day and inter-day RSD were both less than 8%,and the low,middle and high extraction recoveries were within 93.27%~102.65%.CONCLUSION:The method is accurate,sensitive,reproducible,especially not interfered by lovastatin.It's suitable for plasma concentration monitoring and pharmacokinetics study of niacin,especially combined with lovastatin in the clinic.
