丹参注射液对新生鼠缺氧缺血性脑损伤神经细胞的保护作用研究

摘    要:目的:研究丹参注射液对新生鼠缺氧缺血性脑损伤(HIBD)神经细胞的保护作用。方法:复制HIBD动物模型,将60只SD新生鼠随机分为假手术、模型和丹参注射液高、中、低剂量组,在HIBD后48h应用免疫放射分析法测定血清烯醇化酶(NSE)浓度,硫代巴比妥酸法测定血和脑组织丙二醛(MDA)的含量,Western blot检测脑组织转录因子肌细胞增强因子2A(MEF2A)和突触素Ⅰ(Synapsin-1)的蛋白表达量。结果:模型组小鼠HIBD后48h血清NSE、MDA浓度和脑组织MDA含量较假手术组显著升高(P<0.01);丹参注射液治疗后血清NSE、MDA浓度和脑组织MDA含量较生理盐水组明显下降,且呈一定的量效关系(P<0.01)。Western blot检测表明HIBD后48h模型组中MEF2A的蛋白表达量显著高于假手术组(P<0.01);而HIBD后48h模型组Synapsin-1的蛋白表达量显著低于假手术组(P<0.01)。丹参注射液治疗后MEF2A的蛋白表达量明显低于模型组,Synapsin-1的蛋白表达量显著高于模型组,且呈一定的量效关系(P<0.01)。结论:丹参注射液可降低新生鼠缺氧缺血性脑损伤后血清NSE、MDA浓度和脑组织MDA含量,抑制脑损伤后MEF2A基因表达,并抑制Synapsin-1的蛋白表达下调,从而对神经细胞有保护作用。

Protective Effect of Danshen Injection on Hypoxic-ischemic Damage of Cerebral Nerve Cell in Neonatal Rats

Abstract:OBJECTIVE: To study protective effect of Danshen injection on hypoxic-ischemic brain damage (HIBD) in neonatal rats. METHODS: HIBD model was established. Sixty neonatal rats were randomly divided into small-dose, middle-dose and large dose Danshen injection groups,model group and sham operation group. Serum level of neuron specific enolase (NSE) and malondialdehyde (MDA) content of cerebral tissue and serum were tested by immunoradiometric assay (IRMA) and modified thiobarbituric acid method 48 h after modeling. Western blot assay was used to detect the expression of MEF2A and Synapsin-1 proteins. RESULTS: The serum levels of NSE and MDA and the content of MDA in cerebral tissue in model group were significantly increased 48 h after modeling, compared with sham operation group(P〈0.01). The serum levels of NSE and MDA and the content of MDA in cerebral tissue in normal saline group were significantly were decreased markedly after administration in dose-effect dependent manner, compared with model group(P〈0.01). The expression of MEF2A protein in model group was markedly higher than in sham operation group by Western blot assay 48 h after modeling (P〈0.01). The protein expression of Synapsin-1 in model group was significantly lower than in sham operation group 48 h after modeling(P〈0.01). The expression of MEF2A protein in Danshen injection groups was significantly lower than model group after administration while the expression of Synapsin-1 protein was higher than model group in dose-effect dependent manner (P〈0.01). CONCLUSION: Danshen injection can reduce the levels of NSE and MDA of cerebral tissue and serum, inhibit the expression of MEF2A gene and Synapsin-1 protein. It has the protective effect on nerve cells.?
