应用MCS+ED采集自体外周血干细胞治疗糖尿病足的研究

摘    要:目的探讨糖尿病足患者外周血干细胞的采集方法。方法11例患者均给予G—CSF300μg/d动员,在3~5d后用MCS+ED血细胞分离机采集患者自体单个核细胞,处理全血量(4905.88±306.78)ml,用介法将干细胞悬液注入患肢胴动脉。结果采集物中单个核细胞含量(14.83±2.28)×10^9/袋;CD34^+细胞含量(5.32±3.62)×10^7/袋,产品容积(125.38±10.82)ml:10例患者外周血干细胞移植后疼痛、间歇性跛行、患肢发凉、溃疡等临床症状明显改善,1例临床症状未改善。结论MCS+ED采集外周血干细胞安全可靠,效果良好;用介入法进行外周血干细胞移植对患者损伤小、操作方便,可有效改善糖尿病足的症状。

Study on Autologous Peripheral Blood Stem Cell Collected by MCS+ED Therapy for Diabetic Foot

Abstract:Objective The aim of this study was to explore an efficient method collecting peripheral blood stem cells (PBSC) from the patients with diabetic foot for autologous transplantation. Methods Eleven patients were injected intravenously with G-CSF (300 μg/d) for 3 or 4 consecutive days and the apheresis was started on day 4 or day 5 using the MCS +ED blood cell separator. The total volume of apheresis was (4 905.88±306.78) ml. The final collected volume of PBSC was (125.38±10.82) ml containing mononuclear cells (MNC) of (14.83±2.28)×10^6 and CD34^+ cells of (5.32±3.62)×10^7. The PBSC without further manipulation was infused to 11 patients with diabetic foot through the artery of lower ischemic limbs by interventional treatment. Results 10 out of 11 patients had reduced pain, claudication,local cool-feeling and ulcer significant. Only one patient did not have any improvement. Conclusions Diabetic foot patients can be treated efficiently with autologous transplantation of PBSC collected with MCS+ED blood cell separator.
