胃癌细胞产生的一种新型内皮细胞生长因子的分离

人胃癌细胞株 NUGC 2的培养上清液对内皮细胞及纤维细胞有生长刺激作用.收集 NUGC 2的培养上清液,然后经硫酸铵盐析。等电聚焦(IEF)、Mono Q 阴离子交换层析柱、凝胶过滤层析柱(TSK Gel G3000 SW)等方法分离蛋白,得到一种只对内皮细胞有生长刺激作用,对纤维细胞没有生长刺激作用化学不均一的内皮细胞生长因子,其等电点(PI)为2.07～2.90,分子量在45 000～66 000范围,盐洗脱浓度为0.1～0.25M NaCl.它不同于酸性纤维细胞生长因子,也不同于碱性纤维细胞生长因子.

Isolation of a Novel Endothelial Cell Growth Factor Derived from Gastric Cancer Cell

The culture supernatant was found to stimulate endothelial cell and fibroblast proliferation.It was deemed possible that the supernatant contained endothelial cell growth factor the NUGC 2 culture supernatant was collected and purified by salt fractional,isoelectric focusing electrophoresis (IEF),ion exchange chromatography (Mono Q),and gel filteration (TSK Gel G 3000 SW).In the end a chemical heterogeneous growth factor was obtained.The factor only stimulated the prolifera- tion of endothelial cell but not fibroblase cell,its molecule weight was about 45 000-66 000,PI 2.07-2.90,salt eluted concentration 0.1-0.25 M NaCl.
