3-硝基丙酸诱导沙土鼠脑缺血耐受时海马区胶质纤维酸性蛋白的表达

摘    要:目的 观察 3 硝基丙酸 (3 nitropropionicacid ,3 NPA)预处理后缺血再灌注不同时间海马区胶质纤维酸性蛋白(glialfibrillaryacidicprotein ,GFAP)分布、表达的动态变化 ,探讨其与 3 NPA预处理诱导脑缺血耐受的关系。方法 阻断沙土鼠双侧颈总动脉造成前脑缺血模型 ,应用免疫组织化学方法观察星形胶质细胞的反应。结果 GFAP的分布随再灌注时间而变化 ,GFAP阳性星形胶质细胞在海马CA1区主要分布于多形细胞层、辐状层、腔隙层、分子层。 3 NPA预处理后脑缺血再灌注 3dGFAP表达增强 ,7、14d维持在较高水平 ,2 8d时有所下降 ,但与对照组比较仍保持在较高水平。结论  3 NPA预处理诱导脑缺血耐受中 ,GFAP的表达增强保持在较高水平 ,表明星形胶质细胞增生、活化可能与 3 NPA预处理诱导脑缺血耐受有关。

Abstract:Objective To observe glial fibrillary acidic protein(GFAP) distribution and expression in gerbil hippocampus during cerebral ischemic tolerance induced by low dose of 3-nitropropionic acid.Methods Transient forebrain ischemia model was formed by bilateral common carotid artery occlusion.Immunohistochemistry was used to identify astrocyte response.Results The distribution of GFAP positive astrocytes in hippocampal CA1 region changed with different time intervals after reperfusion following ischemia.GFAP-positive astrocytes were found in stratum of multiform cells,stratum lacunosum,stratum radiatum,stratum moleculare and stratum pyramidale of the gerbil after ischemia.The strongest staining was seen on 7th and 14th reperfusion day and the staining intensity remained higher on 28th reperfusion day than the control group.Conclusion It is possible that high level of GFAP expression might be related to neuronal survival in ischemic tolerance induced by low dose of 3-nitropropionic acid.
