探讨参麦注射液对大鼠心肌缺血再灌注损伤中胰岛素抵抗的影响

摘    要:目的:观察参麦注射液对大鼠心肌缺血再灌注损伤中血浆葡萄糖(Glu)、游离脂肪酸(FFA)、胰岛素(Insu-lin)、C肽以及抵抗素(Resistin)水平的影响,探讨参麦注射液对胰岛素抵抗是否有改善作用。方法:取SD雄性大鼠24只随机分为心肌缺血再灌注模型组(MIRI组)、参麦注射液预处理组(SM组)。分组后SM组每天腹腔注射参麦注射液10 mL/kg连续3 d,MIRI组则注射等量的生理盐水。采用冠脉结扎再松开的办法制备大鼠缺血再灌注损伤模型,缺血15min后再灌注45 min。放射免疫法检测血浆Glu、Insulin和C肽的浓度,ELISA法检测血浆Resistin和FFA的含量。结果:与IR组相比,SM组血浆Glu、Insulin、C肽的浓度,血浆FFA和Resistin的含量均明显降低(P<0.05)。结论:SM能够改善大鼠心肌缺血再灌注损伤过程中胰岛素抵抗,对心肌产生一定的保护作用。

Abstract:Objective:To observe the effects of Shenmai Injection on the levels of glucose,free fatty acid(FFA),insulin,C-peptid and resistin in serum in rats with myocardial ischemia reperfusion injury(MIRI).To investigate if Shenmai Injection can against insulin resistance.Methods:Twenty-four heathy male SD rats were randomly divided into two groups(12 for each):ischemia and reperfusion group(IR group),Shenmai Injection preconditioning group(SM group).The ischemia and reperfusion injury models in rats were made by ligating the left anterior coronary artery and then loosening.The content of resistin and FFA in serum were determined by enzyme-linked immunosorbent assay(ELISA);the concentrations of glucose,insulin and C-peptid in serum were measured with radioimmunoassay method.Results:Compared with the IR group,the concentrations of glucose,insulin,C-peptid and the contents of FFA and resistin were significantly lower in SM group.Conclusion:Shenmai Injection can protect myocardium by improving insulin resistance in rats with myocardial ischemia-reperfusion injury.
