摘    要:目的:基于DNA条形码技术,采用ITS2片段对采集至山东烟台的10个桑白皮样品进行基原鉴定研究,以确定样本的基原物种。方法:对采集的桑白皮样品提取DNA,然后进行PCR扩增和测序,应用CodonCode Aligner 2.06(CondonCode Co,USA)软件进行序列拼接及校对,根据GenBank数据库进行BLAST鉴定以及DNA条形码鉴定系统鉴定。结果:DNA条形码鉴定系统鉴定结果显示与系统内物种序列相似度100%,结合GenBank数据库BLAST鉴定结果,所采集的桑白皮样品皆为桑属。结论:DNA条形码技术可作为物种基原的鉴定方法。

Abstract:Objective: Based on DNA barcoding technology, ITS2 fragment was used to conduct identification study on 10 collected samples of Mori Cortex from Yantai, Shandong Province to determine the protospecies of the samples. Methods: DNA was extracted from the collected samples, and then PCR amplification and sequencing were performed. CodonCode Aligner 2. 06(CondonCode Co, USA) software was used to align and calibrate the sequence. BLAST identification and identification of DNA barcoding identification system were performed according to GenBank database. Results: The identification results of the DNA barcode identification system showed 100% similarity to the species sequence within the system.Combining with the BLAST identification results of the GenBank database, we came to the result that the collected white mulberry samples were all of the genus morus. Conclusion:DNA barcoding technology can be used as a method for identification of protospecies.
