FLU-HPLC测定复方吴茱萸巴布膏中阿魏酸血药浓度及药动学研究

摘    要: 目的建立大鼠血浆中阿魏酸的FLU-HPLC测定方法,研究大鼠透皮给药复方吴茱萸巴布膏的药物动力学。方法色谱柱Venusil ASB-Cl8柱(4.6 mm×250 mm,5μm),流动相甲醇-水-冰醋酸(35∶65∶0.6),流速0.8 mL·min-1,激发波长352nm,发散波长467 nm。采用固相萃取法去除血浆中杂质,荧光检测-高效液相色谱法测定了血样中阿魏酸的浓度,并用3P97软件进行了药动学参数的计算。结果方法的最低检测浓度为1.5μg·L-1(S/N=3),绝对回收率范围为87.82%~96.51%。透皮给药后,药-时曲线符合一室模型,主要药动学参数A457.569μg·L-1,Ka 0.146 h-1,Ke0.090 h-1,ρmax 81.581μg·L-1,tmax 8.639 h,Lag time 0.290 h。结论与传统方法比较,本方法灵敏度高、准确,更适用于阿魏酸血药浓度的测定及药动学研究。

Abstract:OBJECTIVE To establish a HPLC method for the determination of FA in rat plasma,and study the pharmacokinetics in rats after transdermal administration of Compound Wuzhuyu Babugao.METHODS The analytic column was Venusil ASB-C18column(4.6 mm×250 mm,5 μm),and the mobile phase was methanol-water-acetic acid(35∶65∶0.6),the flow rate was 0.8 mL·min-1,excitation wavelength was at 352 nm,emission wavelength was at 467 nm.After impurity was removed by SPE method,plasma concentrations of FA were measured by HPLC method,and the pharmacokinetic parameters were calculated by 3P97 software.RESULTS The lowest detection concentration of this method was 1.5 μg·L-1(S/N=3),and the extract recovery was between 87.82% and 96.51% respectively.After transdermal administration to rats,the pharmacokinetic behavior of FA was discribed by an one-compartment model.The main pharmacokinetic parameters were as follows:A was 457.569 μg·L-1,Ka was 0.146 h-1,Ke was 0.090 h-1,ρmax was 81.581 μg·L-1,tmax was 8.639 h,and Lag time was 0.290 h.CONCLUSION Compared with the normal method,the described method in this study is more rapid and accurate,and can be used for the quantitation of FA in rat plasma and for the pharmacokinetic study.
