卵巢癌早期诊断标志物的蛋白质组学筛选

摘    要:目的采用蛋白质组学方法在卵巢癌患者的多种体液中同时筛选出共同异常的多肽,用于卵巢癌早期诊断的研究,并在组织和血清中进行验证,加深对卵巢癌癌变机制的理解。方法采用WCX磁珠结合MALDI-TOF-MS技术在卵巢癌患者的血清、腹水、尿液、囊液中进行比对筛选,将筛选出的异常改变的多肽峰在SWISS-PROT数据库中检索,筛选出相应多肽,并采用免疫组化和ELISA方法在卵巢癌组织和血清中进行验证。结果 4种体液中筛选出来卵巢癌患者血清中下调和上调的多肽峰分别为6081Da和4466Da(P<0.05),在数据库中检索到分别为DLC1和FAK的相应多肽,在卵巢癌组织和血清中呈显著低表达(χ2=29.92,P<0.01)和高表达(χ2=27.65,P<0.01)。结论同一个体的四种体液中同时筛选得到的肿瘤标志物具有较高的特异性,对卵巢癌的早期诊断具有一定价值。

Abstract:Objective To screen the early detection marker for ovarian cancer from multiple kinds of body fluid of patients with ovarian cancer to further understand the mechanism of the carcinogenesis of ovarian cancer.Methods Weak cation beads(WCX) combined with MALDI-TOF-MS were used to screen the ascites,urine,cyst fluid and serum of the same patient(total 30 cases of patients).The candidate peptide peak were retrieved from the data base of SWISS-PROT.IHC and ELISA were used to identify the expression of the candadate protein.Results The candadate peaks screened from the four kinds of body fluid were 6081 and 4466Da which were finally retrieved from SWISS-PROT were DLC1 and FAK.In the demonstration test,DLC1(χ2=29.92,P<0.01) and FAK(χ2=27.65,P<0.01) were identified up-and down-regulated in the tissues and serum of patients with ovarian cancer.Conclusions Biomarkers screened from four kinds of body fluids were highly specific and were valuable for early detection of ovarian cancer to a certain extent.
