同一个体多种体液用于筛选卵巢癌早期诊断标志物的研究

目的采用蛋白质组学方法在卵巢癌患者的多种体液中同时筛选出共同异常的多肽,用于卵巢癌早期诊断的研究,并在卵巢癌患者的血清中进行验证,加深对卵巢癌癌变机制的理解。方法采用WCX磁珠结合MALDI-TOF-MS技术在卵巢癌患者的血清、腹水、尿液、囊液中进行比对筛选,将筛选出的异常改变的多肽峰在训练集和验证集血清中检验其诊断卵巢癌的灵敏度与特异度。结果四种体液中筛选出来卵巢癌患者血清中下调和上调的多肽峰分别为6081Da和4466DaP<0.05),经留一法验证,诊断卵巢癌的特异度为100%,敏感度为100%,盲法检测的诊断特异度为100%,敏感度为87.5%。结论同一个体的四种体液中同时筛选得到的肿瘤标志物具有较高的特异性,对卵巢癌的早期诊断具有一定价值。

Proteomic markers for early detection of ovarian cancer

Objective To screening the early detection marker for ovarian cancer from multiple kinds of body fluid of patients with ovarian cancer to further understand the mechanism of the carcinogenesis of ovarian cancer.Methods Weak cation beads(WCX) combining with MALDI-TOF-MS were used to screen the ascites,urine,cyst fluid and serum of the same patient(total 30 cases of patients).Different peaks were checking in the serum of training set and testing set.Results The candadate peaks screened from the four kinds of body fluid were 6081 and 4466Da.The sensitivity and specificity of the candidate peaks for diagnosis of ovarian cancer were 100% and 100%,by leave-one-off compared with 100% and 87.5% by blinding method.Conclusions Biomarkers screened from four kinds of body fluids have highly specific and valuable for early detection of ovarian cancer to a certain extent.
