紫杉醇、顺铂及康莱特对Lewis肺癌细胞株抑制作用的实验研究

目的:研究紫杉醇、顺铂、康莱特单用及联用对Lewis肺癌细胞株的增殖抑制作用。方法:以不同浓度的紫杉醇、顺铂、康莱特单独及联合处理Lewis肺癌细胞株,24小时后MTT法测定细胞生长抑制作用,应用金氏公式进行联合用药分析。结果:紫杉醇、顺铂、康莱特对Lewis肺癌细胞均有不同程度的增殖抑制作用(P<0.05),但当康莱特的浓度增加到20μl/ml时抑制率不再增加,因此联合用药组中康莱特浓度固定为20μl/ml。作用24小时后紫杉醇、顺铂的IC50分别是15.42、2.59。两种或三种药物联用后对Lewis肺癌细胞株的抑制率显著增加,明显高于单一用药组(P<0.05),其中6μg/ml的紫杉醇与15μl/ml的康莱特联用时,其抑制率接近于12μg/ml的紫杉醇单用药;2μg/ml的顺铂与15μl/ml的康莱特联用时,其抑制率接近于4μg/ml的顺铂单用药,但紫杉醇联用顺铂组与三药联用组的作用差异无显著性(P>0.05)。结论:紫杉醇、顺铂能明显抑制Lewis肺癌细胞的增殖,与康莱特联合用药时可产生较好的协同作用,并且能够减少常规化疗药物的用量。

Study on Anti-proliferation Effect of Paclitaxel, Cisplatin and Kang-Lai-Te on Lewis Lung Cancer Line

Objective:To investigate the growth-inhibition effect of paclitaxel,cisplatin,Kang-Lai-Te and combination of them on Lewis lung cancer cell lines.Methods:Lewis lung cancer lines were treated with different concentrations of paclitaxel,cisplatin,KangLai-Te and combination of them separately.After 24 hours,the inhibitory effects were measured with MTT assay and Jing’s formula was applied to analyze the combination effect of the drugs.Results:paclitaxel,cisplatin,Kang-Lai-Te all showed the anti-proliferation effect on Lewis cancer cell lines in varying degree(P<0.05).However,Kang-Lai-Te’s inhibitiory rate was stagnated when its concentration more than 20μl/ml.Therefore,the concentration of Kang-Lai-Te was set as 20μl/ml.24 hours later,IC50 of paclitaxel was 15.72,IC50 of cisplatin was 2.59.The inhibitory rates of any two or three drugs of them were significantly higher than those in single drug group(P<0.05).The inhibitiory rate of combination 6μg/ml paclitaxel and 15μl/ml Kang-Lai-Te on Lewis lung cancer cell lines was close to that of 12μg/ ml paclitaxel.The inhibitiory rate of combination 2μg/ml cisplatin and 15μl/ml Kang-Lai-Te on Lewis lung cancer cell lines was close to that of 4μg/ml cisplatin.The group of paclitaxel+ cisplatin reached similar cell growth inhibitory rate compared with combination of three drugs(P>0.05).Conclusion:Paclitaxel,cisplatin showed the anti-proliferation effect on Lewis lung cancer cell lines distinctly,and the drugs have better cooperative effect when combinated with Kang-Lai-Te,for the more,the conventional chemotherapy doses can be reduced.
