人巨细胞病毒感染对人涎腺导管上皮细胞存活的影响

目的：研究体内外人巨细胞病毒（HCMV）感染对人涎腺导管上皮细胞存活的影响。方法：用细胞凋亡检测技术研究11例腮腺巨细胞包涵体病（PCID）尸检石蜡包埋组织中细胞凋亡情况；用免疫组化法检测PCID组织存活素、PCNA和P，，的表达；用流式细胞术检测HCMV感染对体外培养的涎腺导管上皮细胞周期的影响。结果：所有包涵体巨细胞凋亡检测呈阴性。包涵体巨细胞存活素和P53，表达呈阴性，其他导管上皮细胞呈弱阳性（±～＋）；75％的包涵体巨细胞核PCNA表达呈弱阳性（±）。与模仿感染组相比，病毒感染组G0／G1期细胞所占百分数在感染36、60和72h显著增高（P均〈0．01）。结论：HCMV感染后涎腺导管上皮细胞并未发生凋亡，而发生了增殖阻滞，大部分细胞处于G1期。

Effect of human cytomegalovirus infection on human salivary gland duct cell

Objective:To evaluate the effects of human cytomegalovirus(HCMV) infection on human salivary gland duct cell(HSGDC) in vivo and in vitro.Methods: Cell apoptosis in paraffin embeded tissues from 11 cases of parotid cytomegalic inclusion disease(PCID) were detected.The expressions of survivin,proliferating cell nuclear antigen(PCNA) and P53 in these tissues were detected by immunohistochemical method.The effects on the cell cycles of HSGDC infected with HCMV and cultured in vitro were investigated by Flow cytometry.Results: Cell apoptosis detection was negative in all of giant cells with inclusion.Survivin and P53 was not expressed in giant cells with inclusion but weakly expressed in other duct epithelium cells.PCNA was weakly expressed in nucleus of 75% giant cells with inclusion.Compared with mock-infection group,cells percentage in G0/G1 stage of virus-infection group showed significant increase at 36,60,72 h post infection(P=0.000,0.008 and 0.000).Conclusion:After HCMV infection,salivary gland duct epithelial cells didn’t develop apoptosis,and the proliferation was blocked in G1 stage.
