胃癌组织中HSP70与Cyclin D1的表达及意义

目的:探讨热休克蛋白70 (heatshock protein 70,HSP70)与细胞周期蛋白D1(Cyclin D1)在胃癌组织中的表达。方法:应用免疫组化SABC 与SP 法检测胃癌、不典型增生、萎缩性胃炎及浅表性胃炎组织中HSP70 与Cyclin D1 的表达。结果:HSP70与Cyclin D1 在浅表性胃炎、萎缩性胃炎、不典型增生和胃癌中的表达阳性率,随病变的进展而升高。HSP70 的表达阳性率分别为42. 9% (12/28)、57. 1%(24/42)、64. 3%(9/19)和77 .1%(37/48),其中萎缩性胃炎、不典型增生及胃癌与浅表性胃炎相比,差异有统计学意义,P<0 .05。Cyclin D1 的表达阳性率分别为7 .1% ( 2/28 )、19 .0% ( 8/48 )、35 .7%(5/14)和52. 1% (25/48),其中不典型增生、胃癌与萎缩性胃炎相比,差异有统计学意义,P<0. 05。大多数受检胃癌组织中,HSP70与Cyclin D1的表达有相关性。结论:HSP70与Cyclin D1 过度表达,在胃癌的产生过程中可能起重要作用。

Expressions of HSP70 and Cyclin D1 in gastric cancer

OBJECTIVE:To study heat-shock protein 70 (HSP70) and Cyclin D1 experssions in the specimens of gastric cancer.METHODS:Immunohistochemical methods (SABC and SP) were used to detect the expressions of HSP70 and Cyclin D1 in specimens of gastric cancer, atypical hyperplasia, atrophic gastritis and superficial gastritis.RESULTS:The positive results of immunostaining for HSP70 and Cyclin D1 protein were observed in superficial gastritis, atorophic gastritis, atypical hyperplasia and gastric cancer.With the development of the disease,the positive rates of HSP70 and Cyclin D1 protein increased. The rates of HSP70 expression were 42.9%(12/28), 57.1%(24/42), 64.3%(9/19) and 77.1%(37/48) in superficial gastritis, atrophic gastritis, atypical hyperplasia and gastric cancer, respectively. Compared with superficial gastritis,the differences of expression rates in all other gastric diseases were significant,P<0.05. The rates of Cyclin D1 expression were 7.1%(2/28), 19.0%(8/48), 35.7%(5/14) and 52.1%(25/48) in above mentioned diseases. Compared with atrophic gastritis, the differences of expression rates in atypical hyperplasia and gastric cancer were significant, P<0.05. In most of the examed gastric cancer tissues, HSP70 expression was correlaled with Cyclin D1 expression. CONCLUSIONS: The excessive expressions of HSP70 and Cyclin D1 may play important roles in the gastric carcinogenesis.
