615系小鼠可移植性乳腺癌株(Ca759)的实验研究

Ca759是1975年9月将615系经产雌鼠的一自发B型乳腺癌经同系移植而建立的一个新瘤株,迄今已传201代,其移植成功率均为100%,皮下移植瘤的潜伏期为5.0±0.3天,存活期为26.9±3.6天。Ca759于常规移植后,局部肿瘤的生长曲线可分为潜伏期、缓慢生长期和快速生长期,即使到晚期局部肿瘤十分巨大,也很少发生溃破,而带瘤宿主也无明显的恶液质表现,转移罕见。随着肿瘤的增长,脾脏也进行性增大,瘤系数与脾系数也均不断增大,两者呈明显的正相关。 Ca759对临床常用十种抗癌药中有8种表现出不同程度的敏感性,特别是环磷酰胺对Ca759的抑瘤率达到100%,对氨甲碟呤、三尖杉酯碱也敏感,这就为抗癌药物的筛选及抗肿瘤作用原理的研究提供有一个较敏感而又有特点的模型。

EXPERIMENTAL STUDY ON A TRANSPLANTABLE MAMMARY ADENOCARCINOMA(Ca759) OF 615 STRAIN MICE

Ca759 is a new transplantable mouse mammary carcinoma model. It has been established since September, 1975 by transplanting a spontaneous type B mammary adenocarcinoma of multiparous female 615 strain mouse into syngeneic recipient mice. Up to now Ca759 has been serially transplanted for 201 generations. The incidence of successful transplantation was 100%, the latent period of the subcutaneous tumor growth was 5.0+0.3 days, and mean survival time of tumor-bearing mice was 26.9+3.6 days. After routine inoculation the growth curve of local tumor might be divided into three periods, namely,the latent,the slowly growing and the rapidly growing periods. Even in the advanced stage the huge tumor mass of Ca759 seldom ulcerated, metastasis was rarely seen and cachexia of tumor-bearing host was not evident. As the tumor grew gradually the spleen of tumor-bearing mice was getting larger,too. And so did the tumor index (g. of tumor/g. of body weight) and spleen index (mg. of spleen/g. of body weight). All of them changed in a significant positive correlation. Ca759 exhibited different sensitivities to 8 of 10 kinds of antitumor agents used frequently in clinical chemotherapy, especially to cyclophosphamide with 100% inhibition rate of tumor weight, and it was also sensitive to methotrexate and harringtonine. Hence the establishment of Ca759 might provide a sensitive model for the screening and study on mechanisms of action of antitumor agents.
