摘 要: | 目的观察氯化锂—匹罗卡品致皮质发育不良(disorders of cortical development,DCDs)大鼠癫痫持续状态(Status epylepticus SE)后行为学和脑电变化特点,探讨皮质发育不良与认知功能和癫痫的关系。方法建立卡莫司汀(BUCN)致皮质发育不良大鼠模型,用氯化锂—匹罗卡品诱导模型鼠SE发作,观察大鼠SE发作前后旷场活动、拒俘反应和水迷宫训练和脑电变化。结果(1)皮质发育不良组(DCDs组)大鼠旷场活动性减少,拒俘反应性增强,水迷宫训练潜伏期延长,与正常对照组(NC组)比较差异有显著性意义(P〈0.05);两组大鼠脑电波均以α、β为主,未见痫性波发放。(2)两组大鼠致痫后认知功能较致痫前均显著降低(P〈0.01-0.05),DCDs组降低较NC组更明显(P〈0.01-0.05);DCDs组大鼠痫性发作潜伏期缩短,SE持续时间延长,与NC组比较差异有显著性意义(P〈0.05);DCDs组大鼠SE后自发性痫性放电较NC组明显增多,波幅增加,组间比较差异有显著性意义(P〈0.01-0.05)。结论DCDs可致大鼠认知功能下降、痫性发作潜伏期缩短和SE时间延长;DCDs大鼠SE后自发性痫性放电明显增强,进一步加重了认知功能的损害。

Abstract: | Objective To observe the the behavior and EEG characteristics in the DCDs rat after Lithium-pilocarpine induced status epilepticus,explore the relationship between DCDs and epilepsy and cognitive function.Methods The modile of DCDs were established by BUCN.With lithium-pilocarpine induced SE in the DCDs rat,to obersve the changs of open-field activity,refusal to prisoners response,water maze training and EEG before and after the onset of SE.Results ①open-field activity reduced,prisoners refused to respond enhanced and the incubation period of water maze training extended were oberved in the DCDs group.The difference was statistically significant between the DCDs group and normal the control group(NC group)(P〈0.05);the EEG in the two groups rat mainly manifested by α and β waves and no epileptic waves were observed.②The cognitive function of the two groups of rat after epileptic seizures were significantly lower compared wite the former,but the DCDs group were significantly lower than the NC group(P〈0.01-0.05);Compared with the NC group,the incubation periodof epileptic seizures in DCDS rats was significantly shortened(P〈0.05);Compared with NC group,the spontaneous epileptic discharges and amplitude after SE were significantly increased(P〈0.01-0.05).Conclusion The DCDs can lead to the cognitive function decreased,seizure latency shortened and the time of SE prolonged;the spontaneous epileptic discharges were markedly enhanced in DCDs rat after SE,and further exacerbating the damage to cognitive function. |
