藻酸盐抗体的制备及其对铜绿假单胞菌生物被膜的影响

摘    要:目的探讨藻酸盐抗体的制备、检定和对院内常见致病菌铜绿假单胞菌生物被膜作用. 方法应用免疫方法制备藻酸盐抗血清;应用脾细胞-骨髓瘤细胞融合技术制备藻酸盐单克隆抗体;应用扫描电镜观察细菌生物被膜形态变化. 结果建立了两株稳定分泌高效价单克隆抗体细胞株;藻酸盐抗体可减少铜绿假单胞菌生物被膜藻酸盐. 结论藻酸盐抗体可中和细菌藻酸盐以防治铜绿假单胞菌生物被膜相关感染.

Abstract:OBJECTIVE To prepare and identify the antibody-serum or monoclonal antibody of alginate, and investigate the protesting role of antibody of alginate against bacterial biofilm (BBF) disease caused by Pseudomonas aeruginosa in nosocomial infection. METHODS Antibody-serum of alginate was acquired by routine immunity method. The spleen cell was obtained from BALB/c mice by programmed immunization and then was fused with myeloma cell line SP2/0. The positive clones were obtained by ELISA method. SEM was used to observe the form of BBF. RESULTS Two kinds of monoclonal antibody of alginate were obtained; P. aeruginosa BBF alginate was decreased by antibody of alginate. CONCLUSIONS The neutralization role of antibody of alginate is one of the mechanism that antibody of alginate prevents and cures P. aeruginosa BBF.
