WT1基因在白血病患者中的表达

目的 研究急性白血病（ＡＬ）患者中ＷＴ１基因的表达情况及其与预后的关系。方法 用逆转录－聚合酶链反应（ＲＴ－ＰＣＲ）的方法，研究ＷＴ１基因在１０２例ＡＬ中的表达。结果 ＲＴ－ＰＣＲ检测ＷＴ１基因的敏感性为１０＾－４。ＷＴ１基因在ＡＬ所有亚型中都有表达，初治和复发的ＡＬ中阳性率为６６．７％，而在５年长期生存者中阳性率只有７．７％（４／５２）。随着完全缓解（ＣＲ）时间的延长，ＷＴ１基因阳性率有所下降，

The expression of WT1 gene in acute leukemia patients

Objectives To determine the expression and significance of WT1 gene in patients with acute leukemia(AL) Method RTPCR was applied to monitor WT1 gene expression in 102 patients with AL Results The sensitivity of RTPCR was 10 -4 WT1 gene could be expressed in all subtype of AL It was detected in 667% of newly diagnosed and relapse patients, but only in 77% of the 5 year longterm survivors WT1 gene expression gradually returned to normal level after CR However, the level increased again before relapse The expression of WT1 gene was associated with the prognosis of AL The CR rate was 458% for patients with positive expression versus 833% for those with negative expression The level of WT1 gene expression was related to the prognosis of AL The relative levels of WT1 gene expression was 0317 in CR patients, whereas it was 0809 in NR patients Conclusions The expression level of WT1 gene was obviously higher in newly diagnosed or relapse patients with AL than that in longterm survivors The level of expression declined progressively in CR period, but it increased before relapse Therefore, WT1 gene is a new prognostic factor and a new marker for the detection of minimal residual disease in AL
