针刺对备用背根节神经元CNTF和PDGF表达的影响

摘    要:目的探讨部分去背根后针刺对备用背根节神经元睫状神经营养因子(CNTF)及血小板源性生长因子(PDGF)表达的影响。方法制备猫双侧备用背根模型(切除双侧L1~L5、L7~S2背根节,保留L6背根节为备用背根节)。针刺位于右侧备用根支配区的两组穴位。针刺7d及14d后取备用背根节用CNTF抗血清(1∶500)及PDGF抗血清(1∶200)行免疫组织化学ABC法染色。分别记数各组阳性总、大及中小神经元的数量。结果正常组左右L6背根节的细胞数无差异,CNTF、PDGF阳性神经元数在7d、14d时与正常组比较无差异。针刺侧CNTF阳性总、大及中小神经元数在针刺7d时均显著多于非针刺侧(P<0.05),而针刺14d时,只有阳性大神经元数多于非针刺侧(P<0.05)。PDGF阳性总及大神经元数针刺7d时多于非针刺侧(P<0.05),针刺14d时,阳性总及中小神经元多于非针刺侧(P<0.05)。结论针刺可促进去背根后备用背根节CNTF、PDGF的表达,这种促进作用显效的时间因不同因子和不同种类的背根节神经元而异。

Abstract:Objective To investigate the expression of ciliary neurotrophic factor(CNTF) and platelet derived growth factor (PDGF) in spared dorsal root ganglion after partial dorsal root rhizotomy and acupuncture in spared root. Methods Fifteen adult cats were divided into three groups. Two groups were subjected to bilateral dorsal root rhizotomy ( L1~L5, L7~S2 DRGs were sectioned, L6 DRG was spared). On the right leg of these operating groups, two sets of acupoints (Zusanli and Xuanzhong; Futu and Sanyinjiao) located in the distribution area of spinal nerve L6 were electro-stimulated alternatively 30 min everyday by electro-needling. The left leg of these group was not treated with acupuncture. On the 7th and 14th day after partial dorsal root rhizotomy and acupuncture, the animals of these groups and another group (intact animals as control) were sacrificed. The spared DRGs (L6) were taken and made into frozen sections and were stained under the same condition. Using specific CNTF antibody (1∶500) and PDGF antibody (1∶200) by ABC immunohistochemistry method. The positive total, large and medium-small sized neurons in spared DRGs were counted. Results Compared with intact group, the number of positive neurons of every factor has no difference at 7th and 14th day. The number of all positive total, large and medium-small sized neurons for CNTF on the acupuncture side increased apparently than those on the non-acupuncture side(P<0.05) at 7th day, but only the rummber of positive large neurons increased at 14th day. The number of positive total and large neurons of PDGF on the acupuncture side increased apparently than those on the non-acupuncture side(P<0.05) at 7th day, while on the 14th day, the number of positive total and medium-small sized neurons increased apparently. Conclusion Acupuncture could promote the expression of neurotrophic factor in spared DRG and this function effected different factor in different period.
