赤雹根总皂苷对类风湿性关节炎大鼠血清OPG/RANKL水平的影响

摘    要:目的:观察赤雹根总皂苷(TSTR)对佐剂性关节炎(AA)模型大鼠血清骨保护素(OPG)、核因子-κB受体活化因子配体(RANKL)表达水平的影响,探讨TSTR治疗类风湿性关节炎的机制.方法:SD大鼠,随机取10只作为空白对照,其余大鼠采用弗氏完全佐剂制备关节炎模型,将造模成功的关节炎大鼠随机分为模型对照组、TSTR 40,80,160 mg·kg-1·d-1剂量组、雷公藤多苷12 mg·(kg·d)-1阳性对照组.各组分别ig给予相应的药物,模型对照组ig等容积的水.连续ig给药21 d后处死.采用ELISA法检测血清OPG,RANKL水平.结果:TSTR各剂量组大鼠血清RANKL水平分别为(98.74 ±10.17),(87.81±17.78),(77.75±14.61) pmol·L-1,均显著低于模型组(120.80±12.14) pmol·L-1(P <0.05或P<0.01);TSTR各剂量组血清OPG水平分别为(71.67 ±2.62),(77.50 ±0.93),(91.04±7.26) pmol·L-1,均显著高于模型组(53.64 ±3.46)pmol·L-1(P<0.05或P<0.0l);TSTR各剂量组OPG/RANKL比值分别为0.71 ±0.08,0.89±0.17,1.13 ±0.24,显著高于模型对照组0.43±0.06(P<0.05或P<0.01).结论:TSTR可能通过影响OPG/RANKL系统而对破骨细胞功能产生影响,从而防治骨破坏.
