溃结灵含药血清对Caco-2细胞损伤模型ITF基因和MUC2蛋白表达的影响

摘    要:目的:观察溃结灵含药血清对人克隆结肠腺癌(Caco-2)细胞损伤模型肠三叶因子(ITF)基因和MUC2蛋白表达的影响.方法:三硝基苯磺酸(TNBS)灌肠法复制溃疡性结肠炎(UC)大鼠模型,采用UC模型大鼠血清刺激Caco-2细胞,造成细胞损伤模型,分别用荧光实时定量-PCR (RT-PCR)法及免疫组化法检测溃结灵含药血清对Caco-2细胞损伤模型ITF mRNA及MUC2蛋白表达的影响.结果:与正常血清组ITF mRNA MUC2蛋白相对表达量(1.00±0.11),(8.62±3.16)比较,模型血清组(1.20±0.16,11.22±2.37),均明显增高(P <0.05,P<0.01);与模型血清组比较,溃结灵含药血清组ITF mRNA(1.76±0.37)及MUC2蛋白表达量(13.94±2.59)均明显增高(P <0.05,P <0.05).结论:溃结灵含药血清可上调Caco-2细胞损伤模型ITF基因和MUC2蛋白表达.

Abstract:Objective: To observe the effect of Kuijieling decoction containing serum on intestinal trefoil factor(ITF) mRNA and mucin 2(MUC2) protein expression in Caco-2 injury model. Method: Three trinitrobenzene sulfonic acid (TNBS) enema was used for replication of ulcerative colitis (UC) rat model. Cell injury model was established by serum of UC model rats, ITF mRNA and MUC2 protein expression were detected by real time fluorescent quantitative-PCR (RT-PCR) and immunohistochemistry. Result: ITF mRNA (1.20±0.16) and MUC2 protein expression levels(11.22±2.37) in model serum group were higher than that in normal group (1.00±0.11,8.62±3.16) (P<0.05,P<0.01); Compared with the model serum group, ITF mRNA(1.76±0.37) and MUC2 protein expression levels (13.94±2.59) in Kuijieling decoction containing serum group increased significantly (P<0.05, P<0.05). Conclusion: Kuijieling decoction containing serum could promote ITF mRNA and MUC2 protein expression in Caco-2 cell injury model.
