肾上腺髓质素对尾加压素Ⅱ刺激的血管平滑肌细胞增殖的影响

目的:观察肾上腺髓质素(ADM)对尾加压素Ⅱ(UⅡ)刺激的血管平滑肌细胞(VSMC)增殖的影响。方法:贴块法培养大鼠胸主动脉VSMC;³H]-胸腺嘧啶(³H]-TdR)掺入测定反映VSMCDNA合成;γ-³²P]-ATP标记的同位素法测定丝裂素活化蛋白激酶(MAPK)活性。结果:UⅡ(10^-8mol/L)显著促进VSMC-TdR掺入和激活MAPK比对照组分别高38%(P＜0.05)和260%(P＜0.01)。UⅡ加10^-10、10^-9、10^-8mol/LADM组VSMC-TdR掺入分别较UⅡ组低7%(P＞0.05)、32%(P＜0.05)和41%(P＜0.01)。MAPK活性分别低24%(P＞0.05)、32%(P＜0.05)和36%(P＜0.05)。结论:肾上腺髓质素抑制UⅡ诱导的VSMC增殖,可能与其抑制MAPK活性有关。

Effects of adrenomedullin on proliferation of vascular smooth muscle cells induced by urotensin Ⅱ

AIM:To observe the effect of adrenomedullin(ADM)on proliferation of vascular smooth muscle cells(VSMC) induced by urotensin Ⅱ(UⅡ). METHODS:DNA synthesis of cultured rat aortic VSMC was measured by ³H]-TdR incorporation. The activities of mitogen activated protein kinase(MAPK) were determined by isotope tagged with γ-³²P]-ATP. RESULTS:UⅡ(10^-8mol/L) significantly increased ³H]-TdR incorporation of VSMC and MAPK activities by 38%(P＜0.05) and 260%(P＜0.01) respectively compared with control group. Compared with UⅡ group, 10^-10,10^-9,10^-8mol/L ADM decreased ³H]-TdR incorporation of VSMC by 7%(P＞0.05), 32%(P＜0.05)and 41%(P＜0.01),respectively, and diminished MAPK activities by 24%(P＞0.05), 32%(P＜0.05)and 36%(P＜0.05),respectively. CONCLUSION:ADM inhibits proliferation of VSMC induced by urotensin Ⅱ through inhibiting MAPK activation.
