芦荟提取物预防大鼠胃缺血再灌注粘膜损伤

摘    要:目的 观察芦荟提取物 (aloeextract ,AE)对大鼠胃缺血再灌注粘膜损伤的预防作用并探讨其机制。方法 用小动脉夹夹闭腹腔动脉 30min ,复灌 1h的方法建立大鼠胃缺血再灌注损伤模型 ,在损伤前给动物灌胃AE[10 0mg/(kg·d)、30 0mg/(kg·d) ]连续 5天 ,每天 2次。测定胃组织和血液中超氧化物歧化酶 (SOD)、丙二醛(MDA)、内皮素 1(ET 1)的含量 ,计算胃粘膜损伤指数 (injuryindex)及胃内 pH值 ,并观察光镜下胃粘膜病理学改变。结果 ①AE可明显减轻损伤模型中的胃粘膜损伤指数 ;②AE能明显降低胃组织和血清中MDA的含量 ,增加胃组织和血清中SOD活力 ;③AE可以降低胃组织和血浆中ET 1的水平 ;④AE对胃内 pH值无明显影响。结论 AE对大鼠胃缺血再灌注粘膜损伤具有预防作用 ,其机制与抗氧化作用、降低胃组织中ET 1,改善微循环等有关。

Abstract:Objective To investigate the prophylactic effect of aloe extract (AE) on acute gastric mucosal injury and to explore its possible mechanism in rats. Methods AE(100 mg,300 mg/(kg·d) was given orally for five days before the acute gastric mucosal injury model was made by ischemia-reperfusion in rats, then the levels of superoxide dismutase (SOD), malondehyde(MDA), endothelin-1 (ET-1) in gastric tissue and blood together with intragastric pH values were measured; injury index was calculated and morphological changes of gastric mucosa were assessed under light microscope. Results ①AE could remarkably decrease injury index and the levels of MDA and ET-1 in gastric tissue and blood. ② AE could significantly increase the activity of SOD in gastric tissue and blood. ③ AE didn't obviously influence intragastric pH values. Conclusion AE can prevent acute gastric mucosal injury caused by ischemia-reperfusion in rats. The mechanism may be related to its antioxidative ability, reduction of ET-1 content in gastric tissue and plasma.
