淫羊藿总黄酮对骨水泥微粒刺激巨噬细胞分泌肿瘤坏死因子的影响

摘    要:目的:探讨淫羊藿总黄酮对人工关节无菌性松动的防治作用机理。方法:运用MTT法检测骨水泥微粒刺激巨噬细胞(macrophage)后的自分泌物对巨噬细胞本身的影响,同时分别应用酶联免疫法及RT-PCR法检测淫羊藿总黄酮对骨水泥微粒刺激巨噬细胞分泌肿瘤坏死因子(Tumor Necrosis Factor-α,TNF-α)水平的影响。结果:骨水泥微粒刺激巨噬细胞的分泌物对巨噬细胞自身有一定的促增殖作用,并且具有一定的剂量依赖性,而RT-PCR及ELISA法则从基因转录、蛋白分泌方面证实淫羊藿总黄酮对骨水泥微粒刺激巨噬细胞具有抑制作用。结论:骨水泥微粒可刺激巨噬细胞TNF-αmRNA的表达并分泌TNF-α,而淫羊藿总黄酮能有效地抑制单核细胞TNF-αmRNA的表达,并通过下调其表达来达到抑制TNF-α的这种分泌,从而对人工关节的松动可能起到防治作用。

Abstract:Objective:To explore the mechanism of total epimedium flavonoids (TEF) on aseptic loosening of joint prosthesis. Methods: Macrophage proliferation under their own secretion induced by PMMA cement particles was measured by MTT assay, and the effect of TEF on tumor necrosis factor-α (TNF-α) mRNA and protein expression in macrophage stimulated by PMMA were measured by RT PCR and ELISA. Results:Macrophage proliferation was enhanced by selfsecretion induced by the particles in a dose-depend manner. The results of ELISA and RT-PCR showed that TEF could inhibit macrophages proliferation stimulated by PMMA on mRNA and protein levels. Conclusion:The PMMA particles can promote the secretion of TNF-α in maerophage cells, and such function can be inhibited by TEF. Thus, TEF might be effective in the prevention and treatment of aseptic loosening of joint prosthesis.
