LC-MS/MS法同时测定人尿液中5-氟尿嘧啶和吉美嘧啶浓度及其尿药排泄研究

摘    要:目的:建立5-氟尿嘧啶和吉美嘧啶尿药浓度的LC-MS/MS同时测定方法,研究胃癌患者口服替吉奥胶囊后其活性代谢物5-氟尿嘧啶和增效剂吉美嘧啶的人体尿药排泄特征.方法:尿液样品酸化后用异丙醇-乙酸乙酯(15:85)提取,采用Hedera ODS-2色谱柱分离,流动相为甲醇-0.01%的乙酸水溶液(35:65);质谱采用电喷雾离子源和负离子多反应离子监测方式.6名胃癌患者单次口服替吉奥胶囊60mg后,测定尿药浓度,评价其尿药排泄特征.结果:5-氟尿嘧啶和吉美嘧啶的最低定量下限分别为0.15μg·mL-1和0.10μg·mL-1;高、中、低浓度的日间与日内精密度均小于15%.6名胃癌患者单次口服替吉奥胶囊后尿液中5-氟尿嘧啶和吉美嘧啶的平均累积排泄率分别为4.9%和18.1%.结论:建立的LC-MS/MS分析方法适用于尿药排泄研究.
