摘    要:目的:探讨人脐静脉内皮细胞株(ECV304)缺氧再给氧(H/R)损伤的发生机制及三七总皂苷(tPNS)对该病理生理过程的影响。方法:将体外培养的ECV304分为七组,正常组;模型组;tPNS高、中、低浓度组;维拉帕米组;吡咯烷二硫氨基甲酸酯(PDTC)组,对各组进行相应处理后,分别检测各组细胞内钙离子浓度Ca2+]i、肌浆网Ca2+-ATP酶活性、一氧化氮(NO)及前列环素(PGI2)含量、细胞活力的变化。结果:模型组、tPNS高、中、低浓度组、维拉帕米组及PDTC组肌浆网Ca2+-ATP酶活性、一氧化氮(NO)及前列环素(PGI2)含量、细胞活力明显低于正常组,Ca2+]i明显高于正常组,其中tPNS高、中、低浓度组、维拉帕米组及PDTC组肌浆网Ca2+-ATP酶活性、一氧化氮(NO)及前列环素(PGI2)含量、细胞活力明显高于H/R组,Ca2+]i明显低于H/R组,且呈浓度依赖性改变,tPNS高浓度组肌浆网Ca2+-ATP酶活性、一氧化氮(NO)及前列环素(PGI2)含量、细胞活力明显高于维拉帕米组及PDTC组,Ca2+]i明显低于维拉帕米组及PDTC组。结论:H/R可激活ECV304,使细胞肌浆网Ca2+-ATP酶活性、一氧化氮(NO)及前列环素(PGI2)含量、细胞活力明显降低,Ca2+]i明显升高;tPNS能有效改善肌浆网Ca2+-ATP酶活性,减轻细胞内钙超载,减轻H/R损伤,且高浓度tPNS效果优于维拉帕米、PDTC。
