兔异体骨髓间充质干细胞穴位与静脉移植对梗死区CD31及bFGF影响的实验研究

摘    要:目的:进行穴位移植与静脉移植两种不同移植途径来观察兔异体骨髓间充质干细胞(MSCs)对梗死区CD31及bFGF表达的影响的研究。方法:将已建立心肌梗死模型的日本大耳白兔随机分为3组,包括模型对照组、穴位注射干细胞组、静脉注射干细胞组。用开胸结扎冠状动脉前降支的方法建立急性心肌梗死模型,进行间充质干细胞分离、培养、传代、流式细胞仪检侧兔MSCs表面标志、收集、BrdU标记、Brdu标记率的检测,利用免疫组化法检测。结果:梗死心肌组织中CD31平均光密度值,与正常组相比较,穴位注射干细胞组均降低,P<0.01,静脉注射干细胞组略升高,P>0.05;与模型对照组比较,穴位注射组P<0.05,静脉注射组P<0.01;穴位注射组与静脉注射组比较,P=0.05。梗死心肌组织中BFGF平均光密度值,与正常组相比较,穴位注射干细胞组升高,P<0.05,静脉注射干细胞组降低,P<0.01;与模型对照组比较,穴位注射组P<0.05,静脉注射组P<0.01;穴位注射组与静脉注射组比较,P<0.01。结论:穴位与静脉注射骨髓间充质干细胞显著促进了梗死心肌组织周围内皮细胞的激活、增殖、迁移,显著改善梗死区血液循环。但与穴位组相比较静脉组CD31没有显著差异。穴位注射骨髓间充质干细胞显著促进了梗死心肌组织中表达促毛细血管新生细胞因子bFGF。
