大鼠坐骨神经损伤后脑源性神经营养因子对脊髓前角内生长相关蛋白GAP-43表达的影响

摘    要:目的探讨大鼠坐骨神经损伤后内源性脑源性神经营养因子(BDNF)对脊髓前角内生长相关蛋白GAP-43表达的调节。方法大鼠坐骨神经压榨损伤后,腹腔注射BDNF抗体中和内源性BDNF,对照组注射生理盐水,动物存活7d或14d,用Western blot与RT-PCR观察GAP-43在脊髓腰骶膨大部前角的表达。结果与对照组相比,注射BDNF抗体后坐骨神经损伤侧脊髓前角内GAP-43蛋白与其mRNA的表达明显下降,上述改变在统计学上均有显著意义(P<0.01)。结论大鼠坐骨神经损伤后内源性BDNF可能参与脊髓前角内GAP-43表达的调节。

Abstract:Objective To investigate the effect of endogenous brain derived neurotrophic factor (BDNF) on GAP-43 expression in the anterior horn of the spinal cord in rats following sciatic nerve injury.
