摘 要: | 目的研究视神经损伤后视网膜的病理形态学变化。方法利用大鼠视神经钳夹伤模型,运用图像分析和形态计量技术对视神经损伤后视网膜中视神经神经元胞体的节细胞(RGCs)核体积密度进行了定量分析,对视网膜各层厚度进行了测量比较。结果①光镜下观察见正常对照组视网膜各层排列整齐而致密,RGC层胞核清晰,各层界限较为清楚,内核层和外核层细胞排列紧密而整齐。损伤后1d可见节细胞层有出血,损伤7d后,节细胞核明显稀疏,外核层及内核层细胞明显减少,核排列不整齐,内网状层亦变薄,视网膜总厚度变薄;②图像分析显示:视神经损伤后7、14、28d视网膜RGCs核体积密度明显下降(P<0.01),分别下降了27.5%、39.8%、49.0%;伤后28d视网膜节细胞层和内网状层厚度明显减少(P<0.05)结论视神经不全损伤导致了视网膜形态结构的变化,伤后7d节细胞明显丢失,28d视网膜出现了萎缩厚度变薄。

The pathomorpholmetry analysis on rat retina following optic nerve crushed |

Abstract: | Objective To analysis the alternation in pathomorphology in retina after rat optic nerve crushed. Methods The model of adult rat optic nerve crushed was established. The retina tissues were fetched at 1,3,7,14 and 28 day post-trauma, and the alternation in pathomorphology of retina was analysed with light microscope. Results (1)In normal retina each layer arranged orderly and densely under light microscope,and retinal ganglion cells(RGCs) nucleus were clear, and border of each floor was rather clarity. Hemorrhage was found in RGCs floor first day after injury.At 7th day, RGCs were evidently lost. Cells of both inner and outer nucleus layer decreased apparently and arranged out of order.Inner reticular layer thinned and thickness of retina became thinner.(2)Image analysis showed: the average volume density of RGCs decreased apparently (P<0.01), the ratio were 27.5%,39.8%and 49.0% separately at 7,14 and 28 days after injury;the thickness of RGCs layer and inner reticular layer became thin evidently at 28th day (P<0.05). Conclusion Changes of morphology were observed in retina after crushed optic nerve. RGCs lost evidently at 7th day after injury, and retina was atrophy and the thickness became thiner at 28th day. |
