| 摘 要： | 目的：分离、培养大鼠骨髓间充质干细胞（BMSCs）,探讨其生物学特性,为组织工程和基因工程提供优势靶细胞。方法：全骨髓细胞贴壁法进行BMSCs的原代培养,倒置相差显微镜观察细胞形态,流式细胞仪检测BMSCs表面抗原,MTT法检测BMSCs增殖能力,并绘制生长曲线。结果：原代培养24h后看见少量贴壁细胞,初起为小圆形,3d后可见梭形类似成纤维细胞,呈漩涡样生长,传3~6代细胞均一性良好,10代以后细胞逐渐有宽大梭形样变,增殖减慢,偶有细胞漂浮、死亡。流式细胞仪检测传3代细胞CD29表达率为92%,CD34几乎无表达。生长曲线显示,传3、6代BMSCs有较强的增殖能力,其中传6代细胞更为明显,传10代细胞的增殖能力较前有所减弱。结论：成功从大鼠骨髓组织分离、培养BMSCs,纯度高,生物学特性符合正常干细胞生长规律,是组织工程和基因工程理想的靶细胞。
