地塞米松致胃肠机能紊乱的动物实验研究

摘    要:目的建立地塞米松致大、小鼠胃肠道功能紊乱的疾病模型。方法以地塞米松ig、sc给药,采用不同给药剂量进行分组实验,观察地塞米松对大鼠胃分泌功能及对小鼠胃肠运动功能的影响,确定建立该模型地塞米松的给药剂量及给药途径,根据模型研究结果选择知柏地黄丸和三九胃泰抑制地塞米松导致的胃肠功能紊乱的试验研究。结果与正常组相比,各组动物ig、sc给予地塞米松后出现小鼠胃肠运动被抑制,大鼠胃蛋白酶活力增加、胃液pH降低、胃壁结合黏液量降低,同时三九胃泰能有效抑制地塞米松导致的胃肠功能紊乱。结论大鼠以地塞米松1.2 mg.kg-1连续灌胃8 d,小鼠以1.0 mg.kg-1连续灌胃5 d,可以建立较为理想的致胃肠道功能紊乱的模型,三九胃泰能有效抑制地塞米松导致的胃肠功能紊乱。

Abstract:Aim To establish the models of gastrointestinal dysfunction caused by dexamethasone(DM) in rats and mice.Methods The different routes of administration and doses of DM groups were studied to determine the feasible experimental condition by the effect of DM on secretion function of stomach in rats and gastrointestinal motility in mice.Results Compared with control group,it was observed that gastrointestinal motility in DM groups were inhibited in mice,the pH value of gastric juice and the amount of gastric wall binding mucus were decreased but the activity of pepsin increased.Conclusions The model of gastrointestinal dysfunction caused by DM is establised successfully at the dose of 1.2 mg·kg-1 for 8 days i.g.in rats and 1.0 mg·kg-1 for 5 days in mice.Sanjiuweitai can effectively inhibit gastrointestinal dysfunction induced by DM.
