丹皮酚对大鼠离体海马神经元NMDA受体的影响

摘    要:目的:研究丹皮酚对大鼠海马神经元缺糖缺氧的保护作用。方法:以原代培养的新生大鼠海马神经元作为研究对象,建立缺糖缺氧损伤模型,倒置相差显微镜下进行一般形态学的动态观察,用MTT法测定神经元存活率。通过放射配基结合实验测定海马NMDA受体结合力变化。结果:丹皮酚可显著降低神经元缺糖缺氧时细胞死亡率,减弱NMDA受体结合力。结论:丹皮酚对离体培养的大鼠海马神经元缺糖缺氧具有保护作用,其作用机制与减弱NMDA受体结合力有关。

Abstract:Objective: To determine if peaonol can protect cultured rat hippocampal neurons from anoxia-induced injury.Methods: We cultivate the rat hippocampal neurons as the object of study and then build the model of oxygen-glucose deprivation.They were randomly divided into 3 groups.Each group was observed by inverted phase contrast microscope;bonding force of NMDA receptors was evaluated by liquid scintillation counting.Results: Compared with OGD group at each designated period,peaonol treatment after OGD obviously increased cell survival rate and reduced the bonding force of NMDA receptors.Conclusion: Peaonol can protect rat neurons from ischemia-reperfusion injury by alleviating morphological damage and increasing neuron viability.
