摘    要:摘要 目的:探索在不同月龄小鼠SAMP8皮层和海马中G蛋白的异常表达情况。方法:以正常老化小鼠SAMR1为对照,利用Western-blot方法检测Gαq/11、Gαi和GαS蛋白在8月龄和18月龄小鼠SAMP8的异常表达。结果:与同月龄SAMR1相比,GαS、Gαi和Gαq蛋白在8月龄小鼠SAMP8皮层和海马组织中的表达无明显异常(p﹥0.05),而到18月龄时,SAMP8皮层部位Gαq表达量明显低于SAMR1(p﹤0.05),海马部位Gαq和 Gαi的表达量也明显低于SAMR1(p﹤0.05)。结论:18月龄SAMP8脑组织中Gαq和 Gαi的异常表达参与了阿尔海默病(Alzheimer’s, AD)的病理进展,并有望作为AD治疗的一个靶点。

Abnormal expression of G protein in the brain of the senescence-accelerated prone mouse(SAMP8)
Abstract:Abstract Objective: To explore abnormal expression of G protein in the cortex and hippocampus of 8-old-month and 18-old-month senescence-accelerated prone mouse(SAMP8). Methods: senescence-accelerated mice resistant 1(SAMR1) as control mice, Detection of Gαq/11, Gαi and GαS protein abnormal expression in 8-month-old and 18-month-old SAMP8 by using Western-blot method. Results: Compared with the month-old SAMR1, expression of GαS, Gαi and Gαq protein in 8-month-old SAMP8 cortex and hippocampus were not different (p> 0.05). Gαq expression in 18-months-old SAMP8 cortex was significantly lower than that of SAMR1 (p <0.05). Gαq and Gαi expression in 18-months-old SAMP8 hippocampus was significantly lower than SAMR1(p <0.05). Conclusion: Abnormal expression of Gαq and Gαi in 18-month-old SAMP8 brain involved in Alzheimer Disease (Alzheimer's, AD) pathological progress and is expected to be a target for AD treatment.
