何首乌提取工艺设计

摘    要:目的 :分别以何首乌提取物 2 ,3 ,4,5-四羟基二苯乙烯-2-O-β-D-葡萄糖苷水平和提取物抗氧化能力为检测指标对何首乌进行提取工艺设计 ,并检验 2 ,3 ,4,5-四羟基二苯乙烯-2-O-β-D-葡萄糖苷和抗氧化能力之间的相关性。方法 :采用正交实验法以乙醇浓度 (A)、溶剂量 (B)、提取时间 (C)和提取次数 (D) 4个因素 ,每个因素选取 3个水平进行实验。结果 :因素A ,D对 2 ,3 ,4,5-四羟基二苯乙烯 -2-O-β-D-葡萄糖苷的含量有显著影响 ,同样对何首乌的抗氧化能力影响也最为显著 ,经检验 2 ,3 ,4,5-四羟基二苯乙烯-2-O-β-D-葡萄糖苷含量与何首乌的抗氧化能力之间呈显著线性相关 (P <0.01)。结论 :以 60 %乙醇 ,8倍量提取 3次 ,每次 1.5h为最佳提取工艺。

Abstract:OBJECTIVE: To optimize the extraction process of root of Polygoni multflori and validate the interrelation between antioxdative capacity and the content of 2,3,4,5-tetrahydroxystilbene-2-O-beta-D-glucoside in root of P. multiflori. METHOD: The optimum extraction was abserved with the orthogonal design; 2,3,4,5-tetrahydroxystilbene-2-O-beta-D-glucoside was determined by HPLC and the antioxidative capacity by using photochemiluminescence detection method with the Photochem supplied by Analytik Jena AG; the concentration of ethanol, amount of ethanol, extraction time and extraction times were the four factors in the extraction. RESULT: The concentration of ethanol and extraction times had significant effect on the content of 2,3,4,5-tetrahydroxystilbene-2-O-beta-D-glucoside and the antioxidative capacity of the crude extract. CONCLUSION: The best extraction process is to extract three times by using 10 fold EtOH(60%), refluxing at 85 degrees C and to extract one and half hours each time.
