摘    要:目的研究Caspase-1在大鼠脑缺血/再灌注损伤中的作用。方法用Longa法制备大鼠大脑中动脉缺血(2h)/再灌注模型,HE染色观察梗死灶的形成,分别用TUNEL染色及免疫组化技术检测鼠脑缺血中心区及半暗带凋亡细胞与Caspase-1的表达。结果在缺血中心区Caspase-1及凋亡细胞主要见于缺血再灌注损伤早期;在缺血半暗带凋亡细胞与Caspase-1于缺血再灌注损伤早期表达不明显,于缺血再灌注24-48h则明显表达。结论细胞凋亡机制参与了缺血后迟发性神经元死亡,Caspase-1参与了其损伤过程。

Abstract:Objective To study the role of caspase-1 in ischemic/reperfusion brain damage of rats. Methods Rat models of the left middle cerebral artery occlusion(2 h)/reperfusion were made using a method of Longa. Infarct zones were confirmed by HE staining. Caspase-1 expression and apoptotic cells in ischemia core and penumbra of brain sections were detected with immunohistochemistry technique and TUNEL respectively. Results Caspase-1 expression and apoptotic cells were mainly detected in early ischemic/reperfusion damage in ischemia core; While in penumbra, they were obviously observed in 24-48 h after reperfusion but not in early time. Conclusions The apoptotic mechanism might involve in delayed neuronal death after cerebral ischemia, and caspase-1 might take part in the damage process.
