环氧化酶-2与基质金属蛋白酶-2在肝细胞癌中的表达及其相关性研究

摘    要:目的探讨环氧化酶-2(cyclooxygenase-2,COX-2)与基质金属蛋白酶-2(matrix metallo-proteinase-2,MMP-2)在肝细胞肝癌(hepatocellular carcinoma,HCC)中表达的相关性和临床意义。方法应用免疫组化SP法检测51例HCC组织、36例癌旁组织COX-2和MMP-2的蛋白表达。结果COX-2在HCC组织、癌旁组织中的阳性表达率分别为80.4%和47.8%(P〈0.05),COX-2的表达与HCC肿瘤大小、静脉浸润以及肿瘤TNM分期相关(P〈0.05);MMP-2在HCC组织、癌旁组织中的阳性表达率分别为68.6%和25.0%(P〈0.05),MMP-2的表达与HCC静脉浸润、肿瘤TNM分期相关(P〈0.05)。COX-2与MMP-2同时阳性表达者25例。结论COX-2和MMP-2在HCC组织中表达上调,在HCC组织中的表达呈正相关(P〈0.01),证实COX-2和MMP-2在HCC发生发展过程中起重要作用。

Expression of COX-2 and MMP-2 in hepatocellular carcinoma and its correlation study

Abstract:Objective To investigate the expression of cyclooxygenase-2 and matrix metalloproteinase-2 in HCC and the relationship between the expression and the clinical pathological parameters.Method Expression of COX-2 and MMP-2 was detected by imuno-histochemical method.Results The COX-2 protein expression rate was 80.4% and 47.8%(P<0.05) in the HCC tissues and paracancerous liver tissues respectively.The expression rate of COX-2 was correlated significantly with tumor size,vein invasion and TNM stage. The MMP-2 protein expression rate was 68. 6% and 25.0% (P 〈0. 05) in the HCC tissues and paracancerous liver tissues respectively. The exression rate of MMP-2 was correlated significantly with vein invasion and TNM stage. A close correlation was showed between the expression of COX-2 and MMP-2 (P 〈 0. 01 ). Conclusions COX-2 and MMP-2 were over-expressed and significantly correlated in HCC tis- sues, which means that COX-2 and MMP-2 might be involved in the process of genesis and progression of HCC.
