消癥散对荷H22移植性肝癌细胞小鼠VEGF、Cyclin-D1表达的影响

摘    要:目的观察消癥散对荷H22移植性肝癌细胞小鼠的抑瘤作用及其机制。方法选择昆明种雄性小白鼠120只,造模成功实体瘤小鼠(60只),腹水瘤小鼠(60只),空白组(10只)。随机将实体瘤小鼠和腹水瘤小鼠分为模型组,消癥散低、中、高剂量组,5-氟脲嘧啶(5-Fu)组,5-Fu+消癥散中剂量组各6组,每组10只。接种H22肝癌细胞24h后按体重给药,每日1次,连续给药10天。实体瘤组小鼠停药24h后,称重并处死动物,剥取瘤块称重,计算肿瘤生长抑制率,采用免疫组化学法检测血管内皮生长因子(VEGF)和细胞周期蛋白D1(Cyclin-D1)表达水平。记录腹水瘤组小鼠生存天数,计算生命延长率。结果消癥散对各组实体瘤小鼠有明显的抑瘤作用,与模型组比较,除消癥散低剂量组外,其余各组差异均有统计学意义(P<0.01);消癥散能够降低瘤组织中VEGF、Cyclin-D1的表达水平,与模型组相比,除消癥散低剂量组外,其余各组差异均有统计学意义(P<0.01);消癥散能够延长腹水瘤小鼠的生存期,与模型组相比,消癥散中剂量组差异有统计学意义(P<0.05),5-Fu组和5-Fu+消癥散中剂量组差异有统计学意义(P<0.01)。结论消癥散对荷H22移植性肝癌细胞小鼠有抑瘤和延长生存期的作用,抑制肿瘤生长的机理可能与下调VEGF、Cyclin-D1的表达水平有关。
