胚胎脊髓神经干细胞的培养和鉴定

摘    要:目的从胚胎脊髓组织中分离、培养并鉴定神经干细胞。方法采用无血清培养和单克隆培养的方法获得大量细胞克隆并对其进行诱导分化,同时应用免疫细胞化学的方法对上述克隆及分化后的细胞进行鉴定。结果从脊髓分离的细胞克隆具有较强的增殖能力,呈Nestin抗原阳性;对其诱导后能分化为神经元、星形胶质细胞和少突胶质细胞。结论所获得的细胞具有自我更新和多向分化的潜能,是脊髓来源的神经干细胞。

Abstract:Objective To isolate,cultivate and identify t he neural stem cells from embryonic s pinal cord.Methods With serum-free cultivation and mono-cloning technology,we acquired much clones and in duced them to differentiation,at th e same time ,immunocytochemistry was used to identify the clones and th e differentiated cells.Results The cells had strong ability to self-renew and showed positive reaction t o Nestin antigen,further more,they c ould differentiate into neurons,astrocytes and oligodendrocytes.Conclusion The cells have the ability to self-renew and are multipotent,whi ch are neural stem cells derived from spinal cord.
