芦丁对单纯性肥胖大鼠血糖和脂代谢紊乱的调节作用

摘    要:目的 探讨芦丁对单纯性肥胖大鼠血糖、脂代谢紊乱的调节作用。方法 34只Wistar大鼠随机分成对照组、模型组和芦丁组。模型组和芦丁组以高能量食物饲喂6周,诱发大鼠肥胖模型,再灌胃给药4周,分批禁食2 0~2 4h ,1 0 %水合氯醛麻醉,右颈静脉及左颈动脉插管,取血测空腹血糖、胰岛素和血脂生化指标,然后进行糖钳实验。结果 模型组空腹血糖、胰岛素、TCH、TG、LDL -C和FFA明显高于对照组(P <0 . 0 5 ) ,芦丁组显著低于模型组(P <0 . 0 5 ) ;糖钳实验显示,模型组葡萄糖灌注速率GIR显著低于对照组(P <0 .0 1 ) ,芦丁GIR显著高于模型组(P <0 . 0 5 )。结论 芦丁具有降低单纯性肥胖大鼠的血糖、调节血脂、改善胰岛素抵抗的作用。

Abstract:OBJECTIVE To study the effect of rutin on blood sugar and lipid metabolism disorder in diet-induced obese rats. METHODS 34 Wistar rats were divided into three groups at random. Control group was fed on normal diet, while both model group and rutin group with the high nourishment diet for 6 weeks, following with saline and rutin for four weeks,respectively. And then the blood lipids, blood sugar, free fatty acid and insulin were determined, the euglycemic clamp technology was taken. RESULTS The blood sugar, insulin, TCH,TG, LDL and the FFA of the model group were obviously higher than control group(P<0.05), while those of the rutin group was obviously lower than the modle group(P<0.05). In the euglycemic clamp study, GIR of the model group was obviously lower than control group(P<0.01), and that of rutin much higher than control group(P<0.05). CONCLUSION Rutin can reduce blood sugar, regulate blood lipids and improve insulin resistance in the diet-induced obese rats.
