大鼠肝缺血再灌注后不同时相ICAM-1分子表达和肝损伤的关系

摘    要:目的:探讨大鼠肝缺血再灌注后不同时相细胞间粘附分子(ICAM-1)表达和肝损伤的关系。方法:Wistar大鼠随机分为假手术组、再灌注30min组、再灌注60min组、再灌注24h组、再灌注72h组共5组;建立大鼠肝缺血再灌注模型,观察不同时相血清丙氨酸转氨酶(ALT)和天冬氨酸转氨酶(AST)水平,以及应用免疫组化方法(SP法)检测肝组织中不同是相ICAM-1分子的表达。结果:大鼠血清ALT和AST水平在再灌注30min升高显著(P<0.05),再灌注60min组达高峰(P<0.01),再灌注24h、72h组血清ALT和AST水平降至正常水平;再灌注后不同时相ICAM-1分子表达水平与血清ALT和AST水平呈显著正相关(P<0.05)。结论:ICAM-1分子参与肝再灌注损伤的过程,其表达的强弱与肝损伤程度密切相关。

Abstract:Objective:To investigate the relationship between the expression of ICAM-1 and the degree of liver injury after liver reperfusion at different time in rats.Methods:Wistar rats were randomly devided into 5 groups:sham-operated group;reperfusion 30min group; reperfusion 60min group; reperfusion 24h group;reperfusion 72h group.The model for reperfusion was established. We observed the level of the aminotransferase in serum and the expression of ICAM-1 in liver tissue after liver reperfusion injury at different time in rats.Results:Compared with sham-operated rats,the level of the aminotransferase in serum after reperfusion 30min, 60min was much higher ( P < 0.05).The level of the ALT and AST in serum is positively correlate with the expression of ICAM-1 ( P <0.05).Conclusion:The degree of ICAM-1 expression is positively correlate with the state of liver reperfusion injury. ICAM-1 takes part in the process of liver ischemia-reperfusion injury.
