补肾中药对骨质疏松大鼠股骨谷氨酸信号的影响

摘    要:目的:探讨补肾活血中药对双侧卵巢摘除引起的骨质疏松大鼠mGluR5的影响。方法:取3月龄SPF级雌性SD大鼠70只,随机分为造模组(60只)行双侧卵巢摘除和假手术组(Sham group,10只)行假手术,术后30 d将造模组动物随机分为6组(10只/组):补肾活血方组(BSHX group)、龟鹿二仙胶组(GLEX group)、二仙汤组(EX group)、仙灵骨葆组药物对照组(XLGB group)及雌二醇药物对照组(EV group)各自按1 mL/100 g体质量给于相应的药物灌胃,假手术组(Sham group)和模型组(Model group)各自灌服等量的生理盐水。连续给药12周后测定各组大鼠血清中Ⅰ型胶原羧基端肽(β-Crosslaps)、总Ⅰ型胶原氨基端延长肽(P1NP)、TRAP(抗酒石酸酸性磷酸酶)的含量;测定大鼠的骨密度;用Western-blot方法检测股骨骨骺Glu、mGluR5蛋白表达。结果:与Sham组比较,Model组大鼠骨密度、血液PINP浓度、Glu表达均显著降低(P<0.01),β-Crosslaps、TRAP浓度及mGluR5表达显著增高(P<0.01)。与Model组比较,各治疗组大鼠骨密度、PINP、Glu蛋白表达显著增高(P<0.05),β-Crosslaps、TRAP浓度及mGluR5表达显著降低(P<0.01),补肾活血组骨密度及Glu表达高于其他各治疗组。结论:补肾活血能有效防止去卵巢模型大鼠骨质疏松的形成,可能通过调节骨内谷氨酸进一步介导骨重建而发挥作用。

Abstract:OBJECTIVE To observe the effect of mGluR5 expression on osteoporosis in ovariectomized rats by kidney tonifying TCM drugs. METHODS Seventy female SD rats of 3 months in SPF were divided into 2 groups randomly, the Sham operation group (n=10), and Model group underwent bilateral ovariectomy (60). After 30 days, the rats of Model group were further divided into 6 groups randomly, the Model group, the estradiol group (EV group), Xianling Gubao capsule group (XLGB group), tortoise deer two fairy glue group (GLEX group), two fairy soup group (EX group), kidney huoxue soup group (BSHX group). Each group had 10 rats receiving corresponding treatments. After 12 weeks, bone mineral density (BMD) of all rats was measured by dual-energy X-ray absorptiometry. Expression of Glu and mGluR5 in left gemur, serum concentration of type Ⅰ collagen carboxy-terminal peptide (β-Crosslaps), the total extension of type Ⅰ collagen amino terminal peptide (PINP), tartrate-resistant acid phosphatase(TRAP) were measured. RESULTS Compared with Sham group, BMD, PINP and expression of Glu of Model group markedly decreased (P<0.01), and β-Crosslaps, TRAP and expression of mGluR5 markedly increased (P<0.01). Compared with model group, BMD, PINP and Glu expression of each treatment significantly increased (P<0.05), β-Crosslaps, TRAP and mGluR5 expression significantly lowered (P<0.01). BMD and Glu expressions of BSHX group were higher than other treatment groups. CONCLUSION The kidney tonifying TCM drugs can prevent the occurrence of osteoporosis in the rats after ovariectomy, which may play a role by regulating Glu in bone to mediate bone reconstruction.
