摘    要:目的:探讨克癌新丸对小鼠肝癌H22移植瘤生长的抑制作用及对荷瘤小鼠免疫的影响。方法:在ICR小鼠右腋皮下注射小鼠肝癌H22细胞,建立小鼠移植性肿瘤模型,通过瘤重计算抑瘤率,观察克癌新丸对H22移植瘤生长的抑制作用;采用腹腔巨噬细胞吞噬试验、淋巴细胞转化试验、Elisa法测定血清细胞因子TNF-α、IL-2、IL-12的水平及利用Western-blot检测瘤组织中IL-2、TNF-α的蛋白表达,观察克癌新丸对H22移植瘤小鼠免疫的影响。结果:在200,400,800 mg· kg-1时克癌新丸对H22的抑瘤率分别为23.12%,38.71%和48.92%,且能提高H22移植瘤小鼠的胸腺和脾指数;克癌新丸能增强H22移植瘤小鼠腹腔巨噬细胞的吞噬功能,促进淋巴细胞转化,提高血清细胞因子TNF-α、IL-2、IL-12的水平,上调瘤组织中IL-2、TNF-α的蛋白表达。结论:克癌新丸明显抑制H22移植瘤的生长,具有较强的体内抗肿瘤活,其机制可能与增强移植瘤小鼠的免疫有关。

Abstract:OBJECTIVE To study the effects of Ke-ai-xin pills on tumor growth and immunity of hepatoma H22xenografts in mice. METHODS Antitumor activity of Ke-ai-xin pills in vivo was observed by the models of implanted H22 in mice. The effect of Ke-ai-xin pills on immune function in tumor bearing mice was investigated by using phagocytosis test and T plymphocyte proliferation test. The expression levels of TNF-α, IL-2 and IL-12 in serum were observed by ELISA assay. The protein expressions of IL-2 and TNF-α in tumor tissues were detected by Western-blot. RESULTS At doses of 200, 400, 800 mg·kg-1, inhibition rates of Ke-ai-xin pills were 23.12%, 38.71% and 48.92% for H22, respectively. Ke-ai-xin pills increased spleen index and chest gland index, improved the phagocytosis function of the macrophage, enhanced the T lymphocyte proliferation, increased the levels of TNF-α, IL-2 and IL-12 in serum, and up-regulated the protein expression of IL-2 and TNF-α. CONCLUSION Ke-ai-xin pills can inhibit the tumor growth of H22 xenografts in mice and exhibit significantly anti-tumor effect in vivo, the mechanism may be related to enhancing immune functions.
