基于HPLC-DAD-Q-TOF-MS的蓝花参化学成分定性研究

摘    要:目的:通过高效液相色谱-飞行时间质谱联用技术(HPLC-DAD-Q-TOF-MS),对蓝花参中化学成分进行分析鉴定。方法:蓝花参醇提物采用SinoChrom ODS-BP C18色谱柱(4.6 mm×250 mm,5 μm),以乙腈-0.5%乙酸水为流动相,梯度洗脱,检测波长为268 nm,流速1 mL·min-1,柱温25 ℃。飞行时间质谱采用电喷雾离子源,在正、负离子模式下扫描采集数据。结果:首次采用液质联用技术对蓝花参进行分析,初步推断出6个化学成分,均为首次发现,分别是紫云英苷、芦丁、槲皮素-7-O-葡萄糖苷、橙皮苷、槲皮素、芹菜苷。结论:建立的蓝花参HPLC-DAD-Q-TOF-MS分析方法简便、快捷、准确,为其进一步研究提供依据。

Abstract:OBJECTIVE To develop a qualitative analytical method using high performance liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry (HPLC-DAD-Q-TOF-MS) for identification of the major constituents in Wahlenbergia marginata (Thunb.) A.DC.METHODS The alcohol extracts from Wahlenbergia marginata (Thunb.) A.DC. were performed on SinoChrom ODS-BP C18colum (4.6 mm×250 mm, 5 μm) with a mobile phase of acetonitrile-0.5% acetic acid in a gradient elution manner. The data were collected in positive and negative ion modes using electron spray with ESI source.RESULTS Based on the retention time and MS spectra, 6 constituents (namely astragalin, rutin, quercetin-7-O-glucopyranoside, apioside, hesperidin, quercetin) were identified by HPLC-DAD-Q-TOF-MS for the first time.CONCLUSION This new approach is applicable for the routine analysis and quality control of Wahlenbergia marginata (Thunb.) A.DC. and provides a constituent data for further in vivo pharmacokinetical studies.
