摘    要:目的 比较纸片扩散(K-B)法和微量肉汤稀释(MIC)法检测18株临床分离白色念珠菌和4株标准白色念珠菌对氟康唑体外药敏试验的检测结果,并分析白色念珠菌对氟康唑的耐药原因。方法 根据K-B法和MIC法检测白色念珠菌对氟康唑体外药敏试验,比较两种方法结果的一致性;通过甲基四氮盐(XTT)减低法测量白色念珠菌生物膜的形成及其与氟康唑耐药的关系。结果 K-B法和MIC法检测白色念珠菌对氟康唑的体外药敏实验结果,经比较二者无明显差异,对氟康唑耐药的白色念珠菌其OD值远远高于对氟康唑敏感的白色念珠菌的OD值。结论 K-B法和MIC法对于检测白色念珠菌对氟康唑的体外药敏试验结果具有较高的一致性;白色念珠菌对氟康唑的耐药与其形成生物膜有重要相关性。

Abstract:Objective The Kirby-Bauer (K-B) method and broth microdilution (MIC) method were used to detect the in vitro susceptibility test of 18 clinical isolates of Candida albicans and 4 Candida albicans against fluconazole, and analyze the causes of Candida albicans resistance to fluconazole.Methods The in vitro susceptibility test of Candida albicans against fluconazole was tested according to K-B method and MIC method, and the consistency of the results of the two methods was compared. The relationship between the formation of Candida albicans biofilm and fluconazole resistance was measured by methyl tetrazolium (2,3-bis-(2-methoxy-4-nitro-5-sulphophenyl)-2H-tetrazolium-5-carboxanilide, XTT) method. Results K-B method and MIC method were used to detect the in vitro susceptibility test results of Candida albicans against fluconazole, and there was no significant difference in statistics.The OD value of fluconazole-resistant Candida albicans was much higher than that of fluconazole-sensitive Candida albicans.Conclusion The K-B method and the MIC method have high consistency for the detection of fluconazole in vitro by Candida albicans. The resistance of Candida albicans to fluconazole is closely related to the formation of biofilm.
