盆炎灵合剂对盆腔炎性疾病大鼠模型的药理作用

摘    要:目的:观察盆炎灵合剂对混合菌液致盆腔炎性疾病大鼠模型的药理作用,探讨其作用机制。方法:36只SD雌性大鼠,随机分为对照组,模型组,妇科千金组,盆炎灵1组,盆炎灵2组、盆炎灵3组,每组6只。采用解脲支原体和致病大肠埃希菌混合菌液感染方法制造大鼠盆腔炎性疾病模型。造模后,妇科千金组(剂量:216 mg·kg-1)、盆炎灵1组(5 mL·kg-1)、盆炎灵2组(10 mL·kg-1)、盆炎灵3组(20 mL·kg-1)每天灌胃给药一次,连续灌胃21 d。对各组别大鼠进行子宫病理学检查,及炎症因子分析。同时对各组大鼠子宫病理切片进行免疫组化试验,检测组织中NF-κB p65和IκB-α蛋白的表达。结果:子宫病理切片结果显示盆炎灵各给药组中性粒细胞和淋巴细胞浸润相较模型组明显减少。与模型组比较,盆炎灵各给药组大鼠子宫组织匀浆中炎症因子的浓度明显降低,且呈明显量效关系。与对照组比较,模型组中NF-κB p65转移到细胞核,给药盆炎灵后,细胞核中的NF-κB p65浓度降低。相对于对照组,模型组大鼠子宫中的IκB-α被降解,给药盆炎灵后,细胞中IκB-α的浓度得到一定程度的恢复。结论:盆炎灵合剂可降低子宫炎性细胞浸润,明显降低炎症因子的浓度,对盆腔炎性疾病具有较好的治疗作用,其作用机制与抑制NF-κB信号通路有关。

Abstract:OBJECTIVE To observe the effects of Penyanling in rats with pelvic inflammatory diseases induced by a mixed bacterial suspension, and investigate the possible mechanism of this effect.METHODS A total of 36 SD female rats were randomly divided into control group, model group, Fukeqianjin group, Penyanling 1 group, Penyanling 2 group, Penyanling 3 group (n=6 in each group). Pelvic inflammatory disease was induced by the mixed bacterial suspension of mycoplasma urealytium and pathogenic Escherichia coli. After model was established, rats in Fukeqianjin group were administrated Fukeqianjin capsules at 216 mg·kg-1·d-1. Rats in Penyanling 1 group, Penyanling 2 group and Penyanling 3 group were intragastrically infused Penyanling mixture at 5, 10,20 mL·kg-1·d-1, respectively. All rats were orally administered for 21 days. Then, the pathological analysis was made on uterine tissues. The expressions of inflammatory factors of all rats were analyzed. Meanwhile, the immunohistochemical test was performed to detect the expressions of NF-κB p65 and IκB-α proteins.RESULTS The uterus pathological section assays showed that the infiltrations of neutrophile granulocytes and lymphocytes in uterus in Penyanling groups were significantly decreased than those in the model group. Compared with the model group, the concentrations of inflammatory factors in each Penyanling group were reduced significantly, and showed a dose-effect relationship. Compared with the model group, more NF-κB p65 protein in model group was transferred to the cell nucleus, after dosing Penyanling, the concentration of NF-κB p65 in cell nucleus was reduced, IκB-α in uterus of model group rats was degraded. After Penyanling administration, the concentration of IκB-α was increased.CONCLUSION Penyanling can reduce the inflammatory cell infiltration in uterus, and significantly reduce the concentrations of inflammatory factors, which suggest effects of Penyanling against pelvic inflammatory diseases. The possible mechanism is associated with inhibition of the NF-κB signaling pathway.
