雄土鳖虫活性物质分离工艺考察

摘    要:目的:采用不同方法对雄土鳖虫虫活性物质进行分离,得到活性组分。方法:比较SephadexG15、SephadexG25对样品的分离情况,需用最佳的物质对雄土鳖虫虫活性物质进行分离,然后再采用高效液相仪对分离后的组分进行鉴别,同时采用半制备液相柱对物质进行分离,获得活性单体。并对活性单体进行体外溶栓和抗凝验证。结果:实验得出SephadexG25对雄土鳖虫虫活性物质分离效果比较好。而后采用高效液相仪对分离后的活性组分进行分析,图谱出现五个含量较大的组分。并采用半制备液相柱进行分别收集后,验证得到第二个组分和第三个组分活性较强。结论:采用不同方法对雄土鳖虫虫活性物质进行系统分离后,得到纯度较高的活性组分,为后期研发和应用奠定基础。

Abstract:OBJECTIVE To separate active substances of the male Eupolyphaga sinensis Walker by using different methods to get the active components.METHODS The sample was separated by using SephadexG15 and SephadexG25.The best material was used to separate active substances of the male Eupolyphaga sinensis Walker,and the separated components were identified by HPLC.The separation of the substance was carried out by the semi HPLC to get active monomer,which was validated for in vitrothrombolysis and anticoagulation.RESULTS The experiment showed that SephadexG25 was good for the separation of the male Eupolyphaga sinensis Walker.Then the active components were analyzed by HPLC.Five mass fractions were developed and collected by the semi HPLC,respectively.The results showed that the second and third components had the best activity.CONCLUSION Active component of high purity can be obtained using different methods from male Eupolyphaga sinensis Walker,which lays the foundation for further development and application.
