亮叶杨桐总黄酮抑制胰岛素抵抗大鼠脂肪细胞信号转导抑制因子3和抵抗素的过表达

摘    要:目的:探讨亮叶杨桐总黄酮对高糖高脂膳食诱导的胰岛素抵抗大鼠脂肪细胞细胞信号转导抑制因子3(SOCS-3)和抵抗素过表达的抑制作用。方法:大鼠随机分为正常对照组、模型对照组和亮叶杨桐总黄酮400、200、100 mg·kg-1·d-1组,首先以高糖高脂膳食饲喂8周建立胰岛素抵抗大鼠模型,随后灌胃给药并继续高糖高脂膳食饲喂4周。于第12周末检测大鼠空腹血糖、胰岛素与抵抗素水平,计算稳态模型胰岛素抵抗指数(HOMA-IRI)的自然对数值,分别以实时荧光RT-PCR与流式细胞术检测脂肪细胞SOCS-3、抵抗素的表达。结果:与正常对照组比较,模型组脂肪细胞SOCS-3、抵抗素过表达,伴空腹血糖、胰岛素与抵抗素水平明显升高。与模型组比较,400,200 mg·kg-1·d-1亮叶杨桐总黄酮组的脂肪细胞SOCS-3、抵抗素表达水平显著下调,伴空腹血糖、胰岛素与抵抗素水平明显降低。结论:亮叶杨桐总黄酮可抑制胰岛素抵抗大鼠脂肪细胞SOCS-3和抵抗素过表达,从而降低空腹血糖、胰岛素与抵抗素水平。

Abstract:OBJECTIVE To investigate the inhibition of adipocyte overexpression of suppressor of cytokine signaling 3 (SOCS-3) and resistin by Adinandra nitida total flavonoids in rats with insulin resistance due to high carbohydrate and high fat diet.METHODS Fifty SD rats were randomly divided into normal control, model control and high, middle, low dose Adinandra nitida total flavonoids groups. The rats with insulin resistance were established with only high carbohydrate and high fat diet for 8 weeks. Then, the high, middle and low dose Adinandra nitida total flavonoids (400, 200 and 100 mg·kg-1·d-1, gavage administration) were administered in the following 4 weeks, and accompanied by the daily high carbohydrate and high fat diet. At the end of the 12th week, the fasting blood glucose, insulin and resistin were detected by ELISA, and then the natural logarithm value of HOMA insulin resistance index (HOMA-IRI) was calculated. The real time RT-PCR was performed to evaluate gene expression of SOCS-3 and resistin in adipocytes, and cytometry analysis was performed for protein expression assay.RESULTS Compared with the normal control group, SOCS-3 and resistin in adipocytes were significantly overexpressed in model control group, and the fasting blood glucose, insulin and resistin were remarkably elevated. The SOCS-3 and resistin expression in adipocytes were markedly down regulated in 400, 200 mg·kg-1·d-1 Adinandra nitida total flavonoids groups as compared with the model control group. Simultaneously, notable decrease was observed for fasting blood glucose, insulin and resistin.CONCLUSION Adinandra nitida total flavonoids can remarkably suppress adipocyte SOCS-3 and resistin overexpression in rats with insulin resistance, thus lower fasting blood glucose, insulin and resistin.
