摘    要:二甲双胍是临床上治疗2型糖尿病的一线药物,主要通过抑制线粒体呼吸链复合物Ⅰ,激动腺苷酸活化蛋白激酶(AMPK),达到减少肝脏葡萄糖输出、降低血糖的目的。然而在临床应用中,二甲双胍的疗效存在明显的个体差异,在诸多的影响因素中,基因多态性是目前研究的热点之一。本文通过文献检索,对近年来二甲双胍疗效与基因多态性关联方面的研究进展进行综述,旨在阐明特定基因对其降糖药效的影响,为预测药物的疗效,实现个体化药物治疗提供参考。

Abstract:Metformin is the first line drug in the treatment of type 2 diabetes. Metformin reduces blood glucose level predominately by inhibiting the mitochondrial respiratory chain complex Ⅰ and the activation of protein kinase (AMPK). In clinical applications, however, there are significant individual differences in the efficacy of metformin. Among the influencing factors, gene polymorphism is one of the hot spots in the recent researches. Through the literature retrieval, the article will summarize the research progress on association between metformin efficacy and gene polymorphism in recent years and it will clarify the effect of specific genes on the hypoglycemic effects of metformin, and provide a reference for the prediction of drug efficacy and the realization of individual drug therapy.
