胰蛋白酶酶切EPO之多肽片段抑制EPO促CFU-E形成的研究

摘    要:目的:观察胰蛋白酶酶切EPO之多肽片段能否抑制EPO促红系形成。方法:对大鼠骨髓进行红系定向体外培养,加入胰蛋白酶酶切EPO之多肽片段干预,观察红系集落形成单位(CFU-E)之数量,判定胰蛋白酶酶切EPO之多肽片段对红系增殖活性的影响。结果:胰蛋白酶酶切EPO之多肽片段加入干预培养后,CFU-E数量明显减少(P<0.01)。结论:胰蛋白酶酶切EPO之多肽产物体外能对抗EPO,抑制红系增殖。其可作为治疗高原红细胞增多症的可能药物进一步研究。

Abstract:Objective:To observe the effects of the hydrolysates of erythropoietin(EPO) on stimulating colony-forming unit-erythropoiesisin in cultured marrows from rats.Methods:Marrows were collected from rats and the mononuclear cells in the marrows were isolated for erythroid oriented culture. Then the level of CFU-E formation was examined under the conditions treated with EPO or EPO hydrolysates of EPO.Results:It was found that the CFU-E formation was significantly suppressed by the hydrolysates of EPO.Conclusion:The hydrolysates of EPO are able to inhibit the activity of EPO stimulating colony-forming unit-erythroid proliferation in vitro.
