正安野木瓜多糖的提取及部分特征鉴定

摘    要:目的从贵州省遵义市正安县产野木瓜中提取并纯化野木瓜多糖(Stauntonia Chinensis Polysaccharides,SCP),对其特征进行初步研究。方法用水提醇沉法提取纯化SCP;苯酚-硫酸法测定野木瓜中多糖含量;红外光谱分析其结构特征;HPLC测定单糖组成;DEAE—Sepharose fast flow分析糖样的电荷分布;Sepharose CL-6B柱层析测定分子量分布。结果lOOg野木瓜粉末通过90℃热水煮提,95%乙醇沉淀得到粗多糖,经重新溶解、Sevag法脱蛋白得到SCP3.40g,得率为3.4%。SCP为米黄色粉末状固体,溶于水,不溶于乙醇、丙酮和乙醚等有机溶剂,其水溶液呈粘稠状,pH弱酸性,Molish反应出现紫色环呈阳性。红外吸收光谱分析结果证明所提取的SCP为含甲酯化糖醛酸的多糖类物质。SCP糖含量为51.6%,其单糖组成主要是半乳糖醛酸,甘露糖,葡萄糖醛酸,比例为46.3:18.5:10.3。同时也检测到少量的葡萄糖,半乳糖和阿拉伯糖。电荷分布测定表明SCP是中性糖和酸性糖的混合物。其分子量接近于58万da。结论SCP为多糖混合物。

Abstract:Objective To extract and purify the polysaccharides from Stauntonia Chinensis (SCP),and to determine the partial characters. Methods Hot water extracting and ethanol precipitating procedure were employed to isolate polysaccharides from fresh Stauntonia Chinensis,after the removal of protein by Sevag method. The purified Stauntonia Chinensis Polysaccharides (SCP)was dried by frozen drier. The amount of total carbohydrates of SCP was measured with phenol-sulfuric acid method, FHR spectrometry was used to determine the characteristic absorption ,HPLC was employed to measure the molecular weight of SCP and the monosaccharides contained in SCP. Results Water-soluble SCP were obtained by a combination of extraction in boiling-water, ethanol precipitation and de-protein by Sevag method. The yield of SCP was 3.4%. Being beige powder, SCP was It was dissolved in distilled water and not dissolved in organic solvent,such as ethanol, acetone, ethyl ether and so on. SCP solved in water is viscous, weak acid and gives a purple ring treated with Molish reagent. Infrared absorption spectrum proved that SCP contains uronic acid methyl ester of the polysaccharide material. SCP was also analyzed by determining the sugar content, uronic acid, molecular weight and charge distribution. The total sugar content of SCP was 51.6% and it was mainly composed of galacturonic acid, mannose, glucuronic acid in the ratio of 46.3: 18.5: 10.3. In addition, minor glucose, galactose and arabinose were determined. According to the distribution of charge, SCP was a mixture of neutral and acid polysaccharides. Although SCP on Sepharose CL-6B showes a little broad peak, it is clear to demonstrate that it contains a main molecule population with a molecular weight of 588.869Kd. Conclusions SCP is compounds of polysaccharides.
