麝香保心丸干预模型大鼠急性心肌梗死后心室重构的机理研究

摘    要:目的:研究麝香保心丸对模型大鼠急性心肌梗死(AMI)后梗死组织病理结构及炎性反应的影响,探讨麝香保心丸抑制AMI后心室重构的机理。方法:通过结扎模型大鼠冠脉左前降支,造成AMI模型。70只SD大鼠随机分为空白组、假手术组、模型组、麝香保心丸等剂量组、麝香保心丸大剂量组及辛伐他汀组。2周后,测定大鼠血流动力学指标、LVWI,并取梗死心肌组织行Masson染色,观察大鼠心肌细胞形态,测定大鼠血浆及梗死组织中IL-6及TNF-α的含量。结果:模型组见大量胶原纤维增生,麝香保心丸与辛伐他汀组大鼠心肌组织中胶原纤维含量较模型组明显减少;模型组LVWI明显增加,与空白对照组相比,模型组LVESP、LV±dp/dtmax显著降低(P<0.01),LVEDP显著增加(P<0.01),血浆与组织IL-6与TNF-α含量显著升高(P<0.01);与模型组比较,麝香保心丸组的LVESP、LV±dp/dtmax显著增加(P<0.05或P<0.01),LVEDP显著下降(P<0.05或P<0.01),血浆与组织IL-6与TNF-α含量显著降低(P<0.05或P<0.01)。结论:麝香保心丸能抑制模型大鼠AMI后胶原纤维的增生,降低左心室重量指数,改善血流动力学指标,抑制炎性反应的发生,这可能是其抑制模型大鼠AMI后心室重构的机制之一。

Abstract:Objective:To observe influence of Heart-Protection Musk Pill(HMP)on the damage of myocardial cell histopathological and inflammatory reaction in rats with Acute Myocardial Infarction(AMI),explore the mechanism of HMP imrove ventricular remodeling.Methods:The left anterior desecending coronary arteries of rats were ligated to made AMI model,70 SD male rats were randomly divided into control group,sham operation group,AMI group,HMP equal dose group,HMP high dose group and simvastatin group.2 weeks later,hemodynamic parameters and LVWI of rats were measured,some ischemia myocardium area were taken to make Masson staining under light microscopy.Then ELISA was used to determine TNF-α and IL-6 in myocardial tissue and plasma.Results:Content of collagen fiber in the myocardial tissue in the infracted and non-infracted areas have been reduced.They had significant difference when compared to model group.LVWI of model group had significant difference when compared with control group.Compared with control group,LVESP and LV±dp/dtmax markedly decreased(P<0.01),while LVEDP markedly increased(P<0.01)and content of IL-6 and TNF-α markedly increased(P<0.05 or P<0.01)in AMI group.Compared with those in AMI group,LVESP and LV±dp/dtmax markedly increased(P<0.05 or P<0.01),while LVEDP and content of IL-6 and TNF-α markedly decreased(P<0.05 or P<0.01)in HMP group.Conclusion:HMP can inhibit hyperplasia of collagen fiber,reduce LVWI,improve hemodynamic parameters and limit inflammatory reaction of rats.This may be one mechanism of HMP improving ventricular remodeling after AMI.
